Your picture just literally saved me money. I was about to buy parafilm because I don't like micropore tape for agar and cling wrap was messy.

But I'll just cut a 2 inch wide piece of my cling off the roll and voila! No mess anymore (ok maybe I'b dumb to not have thought about it sooner but the point is your picture saved me 50$)

Someone here said too an easy way to cut your cling wrap rolls is with a big pair of 3" PVC
cutters

Second attempt at cloning. This time I took an entire baby abort off my APE cake and put it to agar. I saw the mycelium slowly taking over the baby stipe and start spreading. Looks very promising and wondering if I should take a transfer now or wait more?

Hey all!

Here is my thriller: I've zero growth in agar! 6 days after inoculating. Is this normal?

I used the Pastywhyte's Easy Agar Tek, but with another recipe.

Instead, I used:
- 1000ml water
- 20g agar powder
- 16g LME

*Also, I failed in covering the containers with foil in PCing, so I've got heaps of condensation. Could an excess of water in the agar plate prevent spores from growing?
*The spores should be good, as I've just bought them from a trusted vendor.
*Another thing that makes me think if it's an issue or not is the temp variation, ranging from about from 10C - 28C where the agar containers are sitting.
*I did careful sterilisation, SAB, iso, fire...
*Its my first time doing this.

Any idea of what might be happening?

Yeah I agree it just depends. I've had spores germinate and become visible after 3-4 days and I had some plates I had given up on germinate after 12 days. I would give them at least two weeks.

I had some take almost a month to geminate. I was actually going to throw the plate out and saw growth on it. Patience grasshopper.

Water is fine. Not ideal (you avoid pouring hot if possible to reduce condensation) but not a problem. You can drop it if it annoys you. Water moving around on your plate may spread myc and tam.

My petris are lately coming out contaminated by some bacteria or some kind of slime. They come like this out of the PC, because I have them in a sealed plastic bag before using.

Any idea how to stop this?

I use no-pour, and PC 10 petris inside a closed plastic bag (to avoid wasting foil).

My PC has a "pre-steam" feature where it releases some steam from a secondary valve before building full pressure and releasing it through the main escape one (which signal that one has to put the fire at min).  After the main valve starts steaming I lower the fire to min, until the steam stops coming out violently (making the valve jump), and only fizz a bit.  I leave it cooking for 30min, and then let cool down.

After a couple of days I get this thing in them. Mostly close to the walls, or in places where the agar was in contact with water.



Ps. After some research found its probably Bacilus Subtilis. A bacteria that multiplicates via spores, which can resist high temperatures for long periods of time, and that to avoid it, there should be 30+min cooking to kill most of it.

Don't know if a technique similar to the one used to grains, where they're left for a day soaking to activate endospores would work with petris?

Any ideas how to enhance my technique?

I'm having a similar issue with bacteria in my poured glass petris, but not in my disposable plastic petris. 

As I'm still new to glass plates, I'm convinced that I'm not getting the plates fully cleaned/sterilized despite wrapping in foil and using 1h @ 350f in a toaster oven.  Maybe I'm overwrapping them or doing something that's preventing them from getting all the way up to sterilization temps. :shrug:

I left the poured glass plates out in front of my FFU for an hour to set/cool before bagging them upside down.  They were poured at 118f and the plates/lids were still warm (~100f) from being processed in the oven.  I was hoping having the petris be near the same temp would stop the condensation, but it didn't help much at all.
