Re: AGAR ENVY! (Anything and All things agar!) [Re: blazedup]

I slacked a bit on culture work the past couple of months, so pretty much all of these are from fairly dried and very overgrown plates.

Top to bottom left to right

Top : Psilocybe hispanica, transfered Aug 2nd

2 wild Pan cyan, 2 wild Ps cubensis, Aug 4th

Psilocybe subtropicalis/semperviva, Aug 6th

2 Avery's Albino cubensis, 2 Psilocybe natalensis, Aug 6th

Most of these are actually also already on grain or almost ready to fruit and the plates will just go into the fridge.

Ps hispanica and subtropicalis are the only that will be going to grain soon.

Where looks like a good place to take a transfer I was thinking the 9:30 position and 12

I like 9:30 and 12 for now. Probably will be others when it grows a little more too.

I'm back in front of the hood after a good 6 months off.

I'm quietly happy about those 2. Had so much trouble with this RustyWhyte over Xmas just gone but now it seems to want to play ball!

Ya I took transfers from those positions and I’m planning on letting it go for a min to see what else develops.

Any one know what causes this growth everything was fine then a halo showed up and growth slowed down to a crawl.

Sorry, I don't, but I'm curious.. what did you use to take that picture? Is it a microscope, or one of those phone camera attachments?

Fellas, we got growth. I think I have working agar. This was transferred less than 24 hours ago. I’ve never gotten growth like that.

All that remains to be seen is duplicating success. But I am really stoked.

:awegroove: was it the water?  Or what did you do differently?

Noticed this aggressive white contaminant mycelium on a plate in my discard pile. I had not made transfers from it, so I must have put it there because I noticed a satellite colony. Wish I had taken a picture before it turned into this:

Anyone recognize that? I assume it's a kind of mold, but if it's sporulating, then it has white spores. It's interesting that it just grows right over the APE mycelium; I've seen penicillium colonies leave space between themselves and mushroom mycelium on plates. Does trichoderma stay white for this long on a plate?

A bunch of my plates have very aerial growth. Like the transferred wedge fuzzed up...and kept fuzzing. It seems to be a bit fuzzy over the whole growth, not just the center:

Here's the same culture, transferred from the same plate on the same day, but to a plate with the addition of 1% activated charcoal. (All of the pictured plates have 2% MEA, 2% agar-agar, 0.2% nutritional yeast, and used distilled water.) It doesn't have the wide aerial growth; just a little fuzzing where the wedge was dropped:

Another pair exhibiting this excessive fuzzing on MEYA without charcoal:

And another:

Any ideas what is causing this?

I wish I knew for sure. There is debate here, and there is possibly conflicting info in my experience, whether a thin halo is embedded mold cotam or thin shroom mycelium.

I'm getting to the point where I think it can be either.

Transfer away from before that part and maybe use more nutritious agar and see if it changes.

Just add a little more of everything except agar and water in your next batch. Like maybe 5 or 10% more sugars and food, food being malt or potato flakes or grain water.

If it's not contam and it's not genetic shroom issue (throwing up different, thinner, mycelium on an otherwise plate) it might be nutrient based.

You have another issue there too, which is root like mycelium that doesn't quite look like rhizo (strong good, cool looking) mycelium, I've heard someone say that can mean contam.

I welcome any other input here from people with experience. But every time this "halo" discussion comes up there is either no comment, or contradictory comments.

Since growth "growth slowed down to a crawl" I'm thinking invisible contam you can't see. Usually mycelium grows to the edge at a steady rate if temp is the same and no contam.

Did your plates get too hot, like above 90? It's been hot everywhere lately and too much heat can kill / injure a grow and slow it / stop it.

How's the back of the plate look. Can you take a pic? Sometimes the contam is visible from back but not front.

I like the jeweler's loop idea, here are more photo tips:
Taking better Petri dish and shroom grow photos

How hot you growing? I try to keep things at 70 f. Hard in the summer, but over that temp consistently I get more contam.

Remember, mold is fungus, so the mycelium can look similar. Some mold mycelium turns green when it sporulates though. I won't open a dish / jar that turns green. I toss it still sealed.
----
On a related note, I've had a plate that looked like this below and it was absolutely embedded mold:

And a T1 APE clone plate with some weird/random growth that I decided to try and clean up. I took some transfers from around 2-3 o'clock. (to be continued..)

Yah the temps definitely fluctuate from around 85-70 so that and pre mix agar makes me think that maybe it’s temps and nutes (temps fluctuating and high nutes) I’ll transfer and regulate the temp better and see where that goes.

The best (amateur) advice that I can offer is - Trust your transfer(s)

I have a shit ton of plates that are in varied states of growth and stalls and I have to fall back on trusting my transfers.  Here is a classic example

This plate came from a swab and has ridiculously uneven and even wispy growths.  Its even going on month 2 from being put to agar. (Rough, but no visible contams)  And you can even see the spot that I pulled a transfer from.

To this -

These are T3s of that initial plate that have finally found a kumbayah on agar.

Trust the transfer(s)  Some are more successful than others, hence why we usually take more than one spot and go to multiple plates.

Yeah, I have a feeling I may not be able to clean it.. but I'm testing out cleaning different types of contaminated plates on water agar, so I figured I'd give it a go

All the plates streaked from that print have got it.I did manage to get some clean transfers before that popped up.

Just 15ml centrifuge tubes. They are made to be autoclaved so they are PP5 and the lids are meant to have two closed positions. Fully closed and allowing GE. Just leave them quarter turn loose for sterilizing. I get them off amazon, never paid attention to the brand.
