Agar is really easy to mess up but it is also really easy to experiment with ,
Donโ€™t notch it out as a failure but as an experiment.

Some of my favorite dishes have been me half drunk messing around in my kitchen

I had 4 plates left over, I put them in a ziplock.. they should be good to use next time since Ziplocks are sterile, right?

And is the stir plate for LC? Not necessary for agar (nor LC really but a nice touch)

I'm in beast mode tonight.. idk why the hell I waited so long to get my FFU.. it doesn't feel like such a chore to do things like pour plates..


Started some Gandalf, Semperviva, and Hillbilly germ plates.



@guerrilla, yup my plates come in sleeves of 10, it's pretty convenient.

Yeah, I know the plate won't be necessary.. more of a luxury item.. I'll probably use it for both LC and agar

Hey guys any recommendations for getting away from bacteria on agar? I've got 1 strain to work with rn and it's def got some hidden bacteria. I can even see a tiny "droplet" of bacteria on the original spore plate. The recent xfers look better, but still seeing signs of bacteria.

I've tried hot pours, bacteria grew straight through the LME+gentamicin agar? Thought bacteria couldn't grow THROUGH agar, am I wrong? Also did a couple Josex BRF Poke things. I feel like those look better than my agar-agar xfers, but still contamed.

How do you get sneaky, hitchhiking bacteria to f off? Should I try more pokes, hot pours/sandwiches, and just quickly xfer from best looking edges? What's your preferred method, I'm really trying to get something clean to run rn. I have nothing atm :frown:

Try a new print instead of stressing over it.

Yeah when you was talking about often not having a cooling plate handy in a SAB, I was actually going to mention that I really doubt you will have that issue when you have the FFU, as you can pour sooo much more and much easier. I always have countless plates at hand!

Ohh, I was gonna say, I thought you were separating 20 stacks. 10 sleeves would be annoying for me in big pours personally, but as long as they work for you. :smile:

But a stir plate in agar really is pointless, LC I understand, but it's not something that people do for agar, the agar is still going to cool down regardless, and you have a more than reasonable working window so it should not be an issue anyway. The only real reason a stir plate would be implemented in agar is if you had a heated stir plate because you were unable to complete a pouring session in quick succession, for whatever reason. Though I've not once had this issue and doubt you would either. :p

23:57 PC put on heat
00:16 steam begins
00:31 weight added
00:38 psi up to 17 then heat on lowest
00:58 heat killed
01:31 pressure dropped
02:33-02:37 begin pour
02:57 100 plates poured
03:27 plates set

Your times may differ according to how quick you are, your pour temp and your room temp, however, as you can see you have plenty of time for your pour, and when you time your own pours as such, you know exactly when you need to start, and when you have free time to get on with other things, so a stir plate never needs to be in the equation. :thumbup:


EDIT:

Just wanted to add, if you wasnt totally ready to pour after pulling from the PC, a water bath would suffice.

For sure.. I was definitely able to pour a lot faster in front of the hood.. and you're right, I dont see myself being in a situation where I run out of plates again :lol: being able to pour (almost) 80 plates or more in 1 quick pour session changes things.. my agar is probably going to progress much quicker now that I don't need to try and stretch out my plates the way I was before.

Plus, I let them cool in front of the hood while working on other projects, so by the time I went to wrap them up they were all crystal clear.. even the top of my stacks was condensation free.

Some ppl have luck with the sandwich method. Making tiny transfers also helps by using a syringe you flame the needle and when its cooled just try to scrape a tiny piece of mycelium to transfer. Or do this same thing with your scalpel. Dont forget to flame sterilize. You only need a tiny filament of mycelium for it to grow. Good luck!

Recipe for water agar?

I'm just debating on going for Poke - BRF puck, vs sandwich/hot pour.

And how do you guys sandwich? Use the entire other plate, slide scalpel along edges and just try to like flip it on top of the xfer plate? Or just set a square piece of myc over the flat transfer piece? I've tried getting a whole agar plate to fall out directly on top of the other plate, but have had trouble getting the circular piece of agar to release from the plate, even when using a scalpel. When I have managed this, it's been sloppy, had some holes not really and not an "even" sandwich

I'd be tempted to just skip it and start over, but it seems I've got tough luck with getting spores to germinate 100% clean. I always seem to have some bacteria, or mold, riding along with the myc. Super annoying. Like I NEVER see satellite contam colonies. Always just unhealthy looking myc on the germ plate, and subsequent attempts to clean it may look better or worse, but never just CLEAN. Sorry for ranting, if you can't tell I'm massively frustrated w/ this. Legit wondering if I'm cursed and my path in life is just to get screwed by these hidden contams.
