I am too green to be offering advice but both times I have gotten swabs of TW I have had the same issue. Myc looks like a puff ball and unevenly and slowly grows out and even pins on the plate. Something was even on the plins as I could never get it clean. Hopefully somebody has some cleaner spores out there. I would like to have some success with it and hope you do as well.

Me me me over here ya tidalwave have been hard for me lots of lost plates,pf jars,oat jars, and spawned sub to SB.  Once the SB I have flushes out I'm down with tidal wave.

@Danny McBride that isn't a plate pin that's a full grown shrooms.  It's dropped it's spores so like monkey_accident said toss it

Not the most ideal candidate to clone but the best practice would be to rip the mushroom in half lengthways then cut a section from the middle of the stipe (trunk) to transfer to agar.
To open the mushroom to get to the clean contamination free part of the stipe, ripping is better than cutting as prevents pushing any contamination outside of the mushroom into the clean internal you will take the clone from.
Could prove tricky with the little guy you have but nothing ventured nothing gained.

It looks like there is sporelated mold on the plate.. I'd personally cut the plin open to increase chances of a clean sample.

It's not as tricky as markamello suggests though.. just use some sterile Tweezers to rip open the plin.

Yeah I'm replying to the post about the pin on the plate covered in mold. If the pin was from a clean plate no worries transferring it straight to agar but if you took that pin and put it to agar I'd put my money on transferring contam with it.

I think it would be a little tricky only because the pin is so small

Yeah I'm replying to the post about the pin on the plate covered in mold. If the pin was from a clean plate no worries transferring it straight to agar but if you took that pin and put it to agar I'd put my money on transferring contam with it.

Cool yea. I'd take my luck and just transfer away from any contams that come along for a ride. Knowing my luck I'd fk it all up if I tried to take a inner tissue sample from a pin. Doesn't sound fun. I've messed up plenty of times just trying to clone using inner tissue on a mature mushrooms.

I have had successful transfers from plates that have contams on them.  Dont cross over the contam when sampling/transferring from your shitty plate, and work as cleanly as possible.  Heck, Id even opt in for a Josex poke to PF on a nasty plate with some halfway good growths on them if I wanted to experiment or save them.

Does anyone use grain water with other nutrient sources?

I'm looking to try another type agar besides my usual LMEA.

I was reading through some teks and whatnot and saw that one TC recommended using grain soak water diluted while another recommended using it full strength. Then it was mentioned that it was beleived different grains have different strengths of soak water as in oat soak water is much stronger than millet soak water.

I want to use my grain water diluted to increase clearness in plates. My grain water is super dark and cloudy from gypsum. Yes I still use gypsum I have over 100lbs of the stuff so I use it when I remember to add it.

I am thinking about doing 250ml grain soak water 250ml tap water 4gs lme and 8gs agar. Does that sound like it would possibly work or should I try something else. I have currently have access to karo, lme, potatoes, potato flakes, and dog food. I did read that Pasty liked to use grain soak dog food agar so I'm not opposed to trying that either.
