Hey, new cultivator here! I'm wondering if this could be something other than cube myc?

First pic is a Z-strain germ plate from a swab, done on 7-1 (it took about two weeks until one point germinated near the edge, which took over the plate very quickly), and the other two are transfers taken from that on 7-21, and those were fully grown out in 7-10 days. The "puffing up" overlay looking stuff didn't really happen until heavy colonization, it grew fairly uniformly across the surface before that.



A few things concern me: the colonization speed, starting location, and weirdly the smell of the plate. It has an almost tropical-fruity scent, like suntan lotion, bananas, or baby powder or something. But that's just the open plate, I haven't smelled disturbed mycelium directly yet. My other definitely-clean-cube-myc plates definitely smell more neutral.

I really hope this is the myc I'm looking for, but part of me wonders if it's too good to be true. I also know that even if it is cube myc it might not fruit or could have other bad genetics, but I just wanna make sure I'm not inoculating my limited grain jars with mold  xD

Though puffiness can mean contam in agar.

If you read that and post in focus we can tell better.

what temp you working at? I recommend no more than 70 f until you get it. I've been doing 2 years and still try to keep at 70. Though hard in summer sometimes.

On a really hot day I'll open the curtain to my shroom closet to let it get some AC in there, otherwise it can get to 90+, which makes contams outgrow shroom mycelium often.

So I have some nat plate pins I'm putting into lc.  Is there a better note sterile way to do it or just open in SAB cut with scalpel stab and drop into lc is that about it.  Just want to make sure I do it as clean as possible thanks

My camera's busted, already on manual focus, but I opened a plate to try for a better pic.



I'm not worried about underlying contam, as I watched the germ plate like a hawk, and nothing else grew. I swabbed 3 plates, and one did nothing, one only grew whatever this is, and the other has multiple points of germination only recently, no visible contam, which I'll transfer from soon. All of that mycelium is very tomentose and aerial as well.

edit: as for temps, I'm low 70s to mid 80s. Knock on wood, but all of my contam issues have been obvious to spot and easy to fix or mitigate, the heat hasn't been a problem. With this plate, it looked a lot like thin tomentose mycelium as it quickly spread over the surface. It seemed to grow fast but I figured it was just an aggressive colonizer. But then it did all this weird aerial stuff. Maybe you can see, the myc is practically as tall as the agar is deep.

I guess I'm just wondering if anybody else has grown some kind of mold that makes big fluffy pure white clouds over their entire plates, because if not then this is almost surely clean cube myc... unless there's also some kind of completely macroscopically invisible agar contam I haven't read about

Pretty much.. but instead of opening the jar all the way, I'd just crack it open a tiny bit with one hand, drop the sample using the scalpel in the other hand, and close it immediately afterwards.. this also works very well for inoclating jars.

If you have tweezers it's a lot easier to do it smoothly

I wouldn't open a plate to photograph, unless I had already transfered from it. Do you have an SAB? Are you opening and doing work in open room without SAB or flowhood?

Still not in focus, but that's closer.

I think that's contam.

Anyone else want to chime in?

Walk me through what you did to get to this point please. Also, where's a link to your germ plate, if this is a T1 transfer.

Is this Natalias? If so, might be overlay. But if cube, I think contam.

I haven't taken many pics along the way. But my process is really standard, this was just swabs and then transfers in SAB onto 2% agar/2% karo/0.4% potato flake plates. The germ plate was the first out of the three pics in my OP here. This was z-strain cubensis, which I've heard can be an aggressive colonizer, the only thing giving me hope!

I opened the plate in a place where the mold spores can run free if that's what this is. And if it's cubensis mycelium, then opening it isn't gonna do much harm considering the heavy colonization

Hi! I've been lurking for a while, and this is my first post.

I took a bunch of pictures before transferring to new plates (checking whether the cultures perform differently MEYA 2%/0.2%/2% vs the same recipe plus 1% activated charcoal which I have never tried).

Cyclocybe aegerita:


Those are the same culture, but the first ones is on MEYA and the second on MEA. I'm seeing this on all of my cultures - really thin growth on MEA (and a lot more fuzzing up of the donor wedge), but it fills out just fine with 0.2% yeast.

Similar thing with this Golden Teacher agar pin clone (first is undiluted oat water and 2% agar, second is MEA):


I wonder if my malt extract just sucks. It seemed to be ok for the first couple of pours I did. My city water comes from several different reservoirs depending on circumstances, so maybe there's an element present or missing from one of the reservoirs that's made up for by something in yeast and (to a lesser extent) oat soak water?

Having trouble getting any APE clones to grow nicely - they're really slow and tend to grow out asymmetrically. These at least seem to have some nice, if slow, growth.

It was for me. Very rhizo too.

I had a plate of it in a fridge for 7 months. Took a transfer to new plates, this was a week later:


Shrooms I grew from it grew fast, were small but very potent for cubes.


Bruised more green than blue too. The B+ I grew for my first grow did that too.

Sorry for the bad pics, this is my first ever agar batch so the condensation isn't great and makes it hard to focus.



These are gen 1 from MSS. I'm probably going to look to transfer these in a few days, but for the most part the growth is very fuzzy. That seems to be normal from MSS from what I can tell. Should I just choose the least fuzzy looking part to transfer? Or should I choose whatever sector looks the most developed and has grown the furthest?

Also, I have ~8 agar petris that never germinated off MSS, is it ok to transfer to those same petris as long as there's no contamination visible?

Hey y’all. I’ve been poring through posts for about an hour now and haven’t found reliably solid information.

If I were pressure cooking agar for upwards of 40 minutes to an hour at 15-18 psi, could that create problems for me?

Still trying to figure out the hidden variable in my agar prep that causes it to be barren.

You can definitely caramelize the sugars going for longer.. Why are you going that long? 15-20 mins is perfectly fine.

Well I’m not anymore and I think my agar has just started working. Im wondering if there’s a correlation.

I think I was doing a 60 minute cycle. I would have bet you money it was written in a well-rated write up, but I’m going through all the major write ups and they all say 20 minutes. I first started agar in like April, so who knows what I read.

I would really like to pinpoint the exact problem so I can make LME and have clearer plates. Go back to pouring. The BRF appears to be harboring good, fast growth, but it’s opaque.

Anyway thanks for the feedback. 20 minutes might have been what made the difference.

Edit: ah in my write up I did it says I PC everything for 20-30 minutes. I’m not sure that’s right. I’m more confused now lmao
