New platez

I have been deing with a serious amount of bullshit looking plates for the past few months, so this feels pretty great

Those look awesome! I hope to get to this point soon.

Did my first pour with the new hood. I'm surprised how clear they are. They've been sitting for around 5 days and i don't see any contam yet so hopefully they'll be good to go Saturday.

These are tasty too. What recipes are you guys using for those, if you don't mind?

I found the recipe from someone else can't remember who but here it is:

500ml water (i used distilled)
11g agar
7g LME

boil slightly and stir all the clumps out then pc for ~25-30 minutes. This got me 20 plates exactly. I found the tip from D3 to use neon food coloring and it looks great IMO.

I use the same recipe Mr jimmy uses pretty much. I wanna try potato flakes though. 1g of nut. yeast if you wanna get freaky.

I was using neon gel coloring for a long time. One day decided to check out the ingredients. The coloring itself had nutes witch isn't great, but not much of a big deal I don't think. The concerning part were the anti fungal and bacterial conservatives. I don't think that's very advisable.. Brand was Dr. Oetker. Not sure about America, but that shitnis everywhere in Europe.

Thanks for the replies. That's about what I used for my first pour earlier this week, except 10g agar 8g LME. I don't know if the 1g difference for either would change much.

I've got a lot to learn but seeing nice looking plates like these gives me even more motivation. I'm just scratching the surface.

My typical goto these days is
20g agar
16-17g lme
1g nutritional yeast
1000ml distilled water

I wish there was a way to beat the shit out of the yeast enough to make it clear like pure lme.

If you're using 500ml i feel like 1g of agar could actually be almost noticable

Yeah true. Mortar would probably suffice too. I am really paranoid buying any actual lab stuff. It's pretty sketch here. You have to have a business license. I do a little prayer before I order petris. When i was ordering gentamicin i couldn't sleep one night. Lol.

Do you know about Pasty Plates?

It's a tek an admin on here made for using something that's not lab ware for Petri dishes. Won't set off alarm bells.

Is food storage cups or something, I don't know. looks like this:
https://www.shroomery.org/forums/showflat.php/Number/24081330

I've seen a good number of people on here using it. Works but isn't as easy to see in and see what the agar is doing.

Also seen ketchup cups used.

I can find a lot of threads about them but not the original thread.

Anyone know the Pasty Plates original thread?

It's not perfect but if you boil your water and yeast and strain through a coffee filter it helps:rockon:

Badass plates btw

 crack.

I know about pastys  and i hate them with a passion. I hate everything that isn't a petri. I think one needs to realize that business don't voluntary snitch and will make sure not to snitch as it would be very bad for business. Plus buying a 100 dishes every 2 months isn't going to spring any feds into action, i don't think.. But once I can do plates like the ones I posted with a blindfold whilst holding the scalpel between my butt cheeks i might start doing ketchup cups.

I don't think that plates are watched in the US. I don't think the lab supply companies automatically report you to law enforcement for ordering them, like the do with certain drug-making precursor chemicals.

But I've wondered, if someone who isn't working in a recognized lab is ordering a lot of plates and agar, maybe the cops don't think they're growing shroom drugs, maybe they think they're growing bio-weapons.

Just remember, agar is easy, it's sold as food in Asian grocery stores. And some mainstream grocery stores if they have an Asian food aisle.

Doing citizen science now can draw unwanted attention. But it used to be common and accepted. I remember that when I was a kid. My brother got chemistry sets and microscopes when he was 8 and 9 for Christmas. Some chemistry sets before that even had radon and uranium in them. haha.

I was reminded of it when I heard Hamilton Morris say recently that according to things he'd read, in the 50s and 60s, people used to be able to walk into a pharmacists' office and buy ether, beakers, test tubes, and all kids of "dangerous" chemicals.

Apparently back then some Boy Scout merit badges required kids buying some of those things.

Come on, being scared of buying petris because someone is going to think you're growing biological weapons is a level of paranoia even i can't get behind. And i live in a country where the police budget is like 0.0000001 that of America and still have 2 panic attacks a day

Hello - I’m new to agar so hoping for some advice :

I put this bit if mush tissue from my last fruiting into agar to see if it would grow - looking to transfer on to another dish

Which bit should I take ? Or let it grow a bit longer ?

Thanks

Welcome to the wonferful world of seaweed. That's all clean growth. Transfers aren't needed unless you suspect that there is something hitching a ride and especially because that's a clone culture. It's all the same genetics. What did the mushroom that you took the tissue from look like?

I have a question about genetics.

I have a plin (plate pin) clone that I really like. I grew some uniformly good shrooms on a few shoe boxes that all matured at the same time in each box and were stronger than other classic cubes I've grown from MS.

Then I cloned one of the stipes of those clone shrooms, and am now growing out that on SBs.

Do you predict the genetics and potency of the clone of the shroom of a clone will be similar to the original clone?

I know I won't know until I try it, but does anyone here have experience with this?

thank you.

A mushroom is made up of multiple dikaryotic mycelium cells, so say you had a single dikaryotic cell, it would only produce one type of fruit body, essentially a monoculture, it contains 1 organism. Minda 1 organism could be monokaryotic but those don't fruit.
Considering your clone isn't just 1 dikaryotic organism but many, as a mushroom fruit body can contain over a hundred different "organisms", or different dikaryotic myceliums, then cloning any more than 1 cell of a mushroom, an amount invisible to the naked eye, will yield more than 1 "organism", or more than 1 dikaryotic mass which holds its own set of genetic material.

So it will be similar, but far from exact.

We also know that via mitosis, the exchange of nuclei between two dikaryotic myceliums, the bases of di-di mating, that di myceliums will exchange genetic information that can effect the fruit body development, although it is more effective in di-mon or mon-mon.
Like when two monos meet and share their genetic info, two dis will do that as well, and continue to do that. That is the main reason fungi are so phenotypically instable, and why breeding stability into strains is very important.

Hope that makes even just a little bit of since.

I took the tissue from the bigger mushroom here:

I thought he looked like a cheerful chap.

Okay so I want to spread him to another dish but I can do that at a later point then, mostly so I can continue the growth and use for agar-to-grain.  I wasn't sure if it was completely clean or not, so thanks for your input.

On the subject of genetics, this new mycelium growth is a clone of the genetics from the fruit, but would still presumably be a mix (original fruit came from MSS) ?
