appreciate the feedback man!

I'll try to isolate that APE myc from the bacteria - grab a sample directly from the top growth.

Regardless, time for more germ plates

Hey guys, how does this PE plate look to yall? It looks a little weak and sparse to me. How should I proceed?

PE T1 on agar, ingredients consist of:
1 cup water
4.5g agar
3 teaspoons potato flakes
1 drop honey

This is exactly how my PE culture looks  I'm guessing it just be that way sometimes. My first PE jar is like 90% colonized, fingers crossed

Bubles in my agar happens to me every once in a while. IMO, its not a big deal.

I hate my new 1000ml pyrex slim line media bottles because they slosh and splash while I’m pouring making way more bubbles than I’m used to. You know me homie, I’m autistic and there comfort in symmetry and consistency. Things like bubbles or a myc disk off center can ruin my entire mood for the day.

How does that top left plate look to y'all? more tomentose than the rest & the edge from ~10 to 12 o'clock looks a bit weak to me.
the rest imo look fine for grains whatcha think

I realized its going to take a little longer to test each plate that i have ready- they've been sitting out for 1-2 weeks fully colonized. i just read that i shouldn't let my plates colonize to the edges to avoid contam risk but they've all pretty much done that already, so i went ahead and separated my top runners from every set i had and put them in the fridge just now. (wrapped in parafilm, then all together with saran wrap, then in ziploc bag). My initial idea was to let them grow out and sit for a little to see which dishes pin on agar so i could test those first.

how detrimental was my mistake of letting them get to the edge of my dishes? i finally have testers waiting for their turn for grains and i'd hate to have fucked it all up cause i didn't fridge them early.

Minimal risk if you're solid on sterile Tek and clean culture. Still not best practice, but if I'm confident on those other aspects, no worries.

It’s fine. If you want to be extra careful just don’t use any myc from near the edge.

The edge is closest to the air so it’s easier for something to land there, but pretty much everything is obvious on agar. So if the edge looked clean a few days after you last opened the plate it should still be fine.

Has anyone ever had germ plates come out like the AA+ I have germinating here? It looks super concentrated and don’t know if this is looking right…

Also has anyone transfer what seemed to be straight rhizomorphic growth on to a plate and the plate comes on straight tomentos?

The aa+ is bacteria.

Myc can switch between growth types based on how the plates are made.

that AA on the left is some type of bacteria/mold no doubt. thats not myc
and yeah, myc can change from rhizo to tomentose and back and forth forever depending on conditions and agar medium recipe changes

edit: lol AKA beat me to it :whathesaid:

If I remember correctly some guys were talking about adding nutritional yeast for more rhizomorphic growth
never tested it myself but worth a shot if you like the rhizo look
