The Hyphal Knot
&
Rhizo Transfer Pictorial


This is a little write-up on a couple techniques that I like to use when making transfers

This first series of pictures goes through the invitro hyphal knot transfer. I simply use a stainless steel dentistry pick to pluck a hyphal knot off the plate and tranfer it to a new plate. Make sure that the tip of the dental pick is completely cool before plucking it off the mycelium. Otherwise, it will stick and be almost impossible to get off. You also want to make sure that when you transfer to the new plate that only what has been flamed can hover over the open plate.


As you can see, the hyphal knot is so small you can barely see it on the tip of the pick.


The first pic below was taken just after I wrapped the plate. The second pic was taken today just 5 days after the transfer.



This second series of pictures demonstrates the transfer of a single thick, ropey strand of mycelium. I simply press the curved part of the pick into the agar across the rhizomorphic strand of mycelium in order to separate it from the rest of the culture. Then I use the tip of the pick to pick up the strand to make the transfer. Again, you want to make sure that when you transfer to the new plate that only what has been flamed can hover over the open plate.

Very nice :thumbup: Glad you cud find a homie to take the photos. Them are some tiny tissue samples.

Do you notice any difference in growth or fruiting between rice sized transfer and atomic scale transfer?

My wife is the best.

The only difference in growth that I've seen in these types of transfers is that they don't really have a recovery phase. But overall, colonization is just the same as any other transfer that I have made. I don't typically make large transfers so I don't have any opinion on that.

As far as fruiting goes, I'm not sure how much of a difference it makes. I believe that in an exceptional fruiting environment all or atleast most genetics will fruit just fine.

The advantages of this technique which separate it from a standard transfer are that this technique uses the tiniest bit of mycellium. The smaller the transfer, the less chance of carying something else with it. It also eliminates the transfer of agar. I believe the agar itself along with the 3 or 4 cuts made to take the transfer cause lag time after the transfer. There is virtually no lag phase after this type of transfer.

Edit: I also believe that the hyphal knot transfer does give a clear indication that the culture will fruit. But like I stated previously, I believe all or atleast most genetics will fruit just fine in an exceptional environment.

Indeed. Very cool. I'd guess that the cutting does the same thing as a shake and causes the myc to draw back for a day or three. Seems that with this tek your are removing myc without causing it to draw back. Hmmmmmmmmmmm. Not that a few days really matters to me.... Most of the time. Thanks for the new thought. I love those.

This thread has been completely derailed. Thank you. If you don't agree with something, you do have the ability, maybe not the self control, but the ability to just ignore it. Some folks just feel the need to have the last word. It is a sad sight to see such an ego forcing itself to defend itself for no real reason.

Now, to get back on topic. If nothing else, the pictures provided demonstrate good sterile technique and great visuals that can help those who are new to cultivation and agar work. You can never have enough good info and visuals for others to learn.
