I’ve had a couple swabs that I streaked a bunch of plates and got nothing until I cut the whole handle off and jammed the swab into the agar.

Some people do the smarter version and tweezer some threads off to stick in the plate but I couldn’t get any to come loose.

Hey man, I don't know which ones are best to harvest, but if you find out, can you tell me? I have some at the same stage by the look of it 10 days.

My plan was to just start picking out bits that start looking ropey and grain spawn from that but not sure now. what do you know?

So this is an all out attempt to reduce the rate of auto-contams running about 30-40% currently.
Just say no to immaculate contamination!

9/9 Wednesday - Plates, Corning Borosilicate 100x15mm
20 plates in foil oven sterilized, 400 F, 2 hrs. ... should be 450 F
Cool down in oven 2 hrs
Carry in shoeboxes to SAB

9/10 9:00 Thursday - “Team Telephone” Agar
8g 1.6% Agar, 7.5g 1.5% Pilsner LME, green, no broth
Soak LME in 500mL bottled water before - 1 hr
Agar 20 min PC, took at least 20 to cool till pressure lock clicked.
Then opened the PC and tightened the media cap with foil still on.
Then off to the SAB to cool to 120F, this time.
Washed 90 Qrt SAB, rack, and table, w/Clorox & towel
Books on top of SAB to anchor it down and reduce effects of bumping.
Wet towel under 3” high rack. Foil wrapped plates in SAB.
Pour in 2 hrs.
What a pain taking the foil off in the SAB
Media bottle ring fell off, but only a couple drips.
I marked the drip spots on each plate with a Sharpie.
Maybe 200mL left over, some extra plates on top, to reduce condensation, of 4 stacks of 5 ea. 11:00
Oooh, really clear green!
Not going to let them sit too long before wrapping stacks of 5.
Trying to avoid auto-contam, immaculate contam.
But if there’s too much condensation,
then I’ll try putting a plate sleeve on them and cool next time...
Also this time, I have a sheet over the SAB,
because I might have a visitor.
12:00 wrapped! That’s a wrap!    :awedance:

Hey. So I’m on my second transfer of PE. I cut a large wedge of some rhyzomorphic myc which has continued to grow but it’s hovering over the new agar. My question is would you guys cut the wedge into thirds while the myc is strong and place onto new plates or just let it keep growing out until it hits the plate on its own. I know it was a large transfer but I’m still not great with the scalpel and I’m paranoid about contams so I was working quickly. I’m working on purging my agar thinner so this doesn’t happen again.

Another question. Do you guys transfer as soon as you think you have contamination on the plate or do you wait until the myc grows out but is still away from contamination. The plate has a spot of bacteria but it’s small so far. Thanks!

I would transfer in 2 or 3 days personally.

Mold i try to transfer away from asap. But bacterial sattelites usually stay put

Depends what the contam is.

Mycelium generally can "outrun" bacteria efficienctly enough that waiting for it to grow out more will work well.

If it's thought to be contaminated with some type of sporulating mold...well then transfering ASAP is ideal.



So there are different answers I guess depending on what you're dealing with. Hopefully that provides some insight. Surely some other members have advice on this too.


I'm working on a RW clone culture but the mycelium is looking iffy. I think it might be the formula I've been using for agar as of late but IDK. Might try to whip up a batch with some grain soak water, see if I can get better looking growth :confused:

It's a seriously kick ass clone so, I reallly don't want to give up. Worried it has some hidden mold or something but who knows. The investigation continues

first run was a little bumpy... er... chunky... but poured some decent plates and learned a lot along the way, pumped to get something growing on these ^_^
