What's a good contamination rate for your poured agar plates?
I've recently been pouring a lot of plates and find about 1 of every 20 (sample size of 100) gets some satellite colony on them while they're waiting patiently to receive a transfer.
Using a SAB. Is this normal or is there something in my sterile technique that I should be addressing?

I shoot for 100% sterile. 1 in 20 is a no no imo:nono:

Do you notice a pattern? Sometimes my bottom plate contaminates because I'm probably not as careful as I need to be while hovering the stack during the pour.

This tells me I'm doing a shitty job:tongue:

Me too.

I started pouring the first three and then adding the rest of the stack on top of those and pouring from there.

It always feels too awkward lifting the whole pile to do the first one or two plates.


Besides the occasional problem with the first plates, usually bacteria under the agar, I rarely see contamination on the rest.

I'm extremely careless and clumsy in my SAB. Sometimes I drop a plate, or take a cover off on accident when removing them from the sleeve but kinda figured everyone had these problems.

I haven't had ANY contamination in my transfers though (sample size of 50), so it leads me to think I'm doing something wrong when pouring plates or maybe a QC thing with the celltreat 100mm's. I'm even sloppier when taking transfers, fumbling around with parafilm and a hot scalpel...

Not sure I’ve had any pour plates contam... not that I can recall at least. I work in front of a hood though. I can be super sloppy and get away with it.

I work with only 5 stacks, but the glass ones are heavy!
Anyway, ya the bottom plates are getting contam.

Maybe did better last time, nothing yet.
I'm wiping the SAB,rack, and all with a towel of Clorox and then letting it
sit an hour. All plates get wiped, scalpel, gloves, clorox towel goes under the rack.
No bumping the SAB, slow movements,

Lol, I'd be happy just to get one clean plate. Since moving over to pours I've had nothing but shit luck. Think I'm just gonna stick to my pre pours and see where that gets me.

If you can do transfers then pouring plates should be no problem. I’ll get tons of transfer contams but almost never while pouring.

That's whats freaking me out, Aka.
I'm sure I'm missing a step along the way and go "eureka" as soon as I realize it, but for the meantime I have no clue, and was seeing if it was something anyone else experiences.

I leave my plates unwrapped in the SAB for a day or so in a room with slightly moving air and arm holes covered with a towel to reduce condensation. I'm thinking this is the weakest point in my technique, but I've seen it suggested multiple times.

Could also be something dropping off my hands. With transfers I wipe my hands with alcohol every couple plates, but when pouring I do 20 without even taking them out. Could definitely invite a hitchhiker or stir something up in that time.

Edit: on that topic, does anyone else feel that final destination moment when you take your scalpel to a flame with some residual alcohol on their hands...?

lol yeah or if I’m impatient and just wiped down the scalpel too. So far no hand fires.


Are you using plates? It could be from the lids lifting a little bit while taking them out or moving them around.

The worst is undercooking the agar and getting bacteria in every plate.

I'm looking for a stainless steel straw (preferably locally) for them perfectly round transfers.

I plan on taping my scalpel to the other end so I can hold em both with ease

I can contribute to this.


Tonight I thought I was going to drop a wedge to my first grain jar. Had a nice sized puck ready to go and it fell off the scalpel and landed face down on a wet towel. FML. Looks like I'm starting grain in about another week and a half or so if I don't drop the damn thing again.

I made one of those about a week ago. Found the straws at my local grocery store.

I was gonna say I’ve been seeing metal straws all over the grocery stores here. Just in random spots where they’ll hang some small shit or little toys from the shelves.

And the camping section.

Yup. After watching D3's thread a couple weeks ago and not being the type to wait on ordering a punch set online, I started looking for alternatives.

Found mine at Target off the bay end of the camping section on sale for like $4 for two straws with silicone bendies on them. I wanted something like Dr. Mario's with the actual curved steel part, but either were pricey or didn't seem like the size I wanted. $4 seemed worth the risk of failure. lol

Usually the kitchen section or camping sections will have a variety, as well as the bay ends or impulse buy gauntlets near the registers of stores.

The one criteria I was making sure to check was the material that they are made of. The straws I got were 18/8 Food Grade Stainless Steel. I didn't want to pick something made of some strange alloy that'd melt or get weird on me.

My knife skills aren't that great yet, and having a perfect puck you could slip your blade into made it easier, pick up a piece and gently flip it over onto a plate with less chance of fumbling it.

Made a transfer tool using a brass 223 casing and old scalpel. Cut a hole in the side so i can pop the transfer out cut the base and crimped it onto a old scalpel.
