It pretty much did just germinate. Does look weird though, huh? Out of the 18 plates I poured only one is showing contams that I can identify. Looks like yeast all around the edge of the plate.



One thing I did notice though is when I made the first 8 plates I thought I really screwed it up. My sterile tek wasn't on point and I didnt let the agar cool for long enough. When I swabbed with spores, the loop went deep inside the agar versus across the top.

In thinking that I completely messed up, I made another 10 plates the next day. This time I was very careful. Sterile tek was really good, the agar cooled enough to not cause a ton of condensation and the loop swabbed across the top of the agar leaving a trail of spores in a nice zig-zag pattern.

The first 8 plates (that I thought had no hope) germinated 3 days after inoculation and are currently developing nice spots of mycelium.



Its been 4 days since the last 10 plates were inoculated and they havent germinated or began to show growth of any kind.

Do spores germinate faster inside of the agar then they do on the top? Its the same print and same recipe the only difference is that I did everything a lot better.

Just got done doing what is a lot of agar work for me. Got some AA and Natal on agar now and did a T5 on a pleurotus citrinopileatus culture im training to eat a stump. Idk how well it's going to work but might as well try something

They prob did like it in the agar. Super wet with lots of nutes.

Myc is crazy resilient, I had a plate dry up to the point it looked like a corn chip just all curled up and crispy. I broke a piece of and put it on a new plate and it grew fine.

With spores Iโ€™ve almost always seen germination by day 5 or not at all. I usually leave them for a month just to see and maybe once or twice some grew between 7-10 days.

Any suggestions on where to take transfers from these germ plates?
Is it better to go larger or smaller than the indicated triangles?

Personally I try to go as small as possible.

The last plate looks like a good plan. The ksss I donโ€™t like the top spot but the bottom looks good. First plate I just donโ€™t like any of it so far.

I streaked 5 germ plates with an APE swab and only got one germ point after 2 weeks. . .
Does that mean that the colony is pretty much an isolate?

T1 plates are awful regular growth. . .

Sorry for reposting, very interested in an answer. . .

Can someone give me the run down of how people cross varieties  ? Can you cross any two cubes together?

Ecuador, Costa rican, Treasure Coast MS Syringes to PDA using an inoculation loop drawing a Z. I've seen many pictures of below and above agar plates grown out but I've searched and haven't found any taken from the side, anyone with pictures to show of mycelium growth when first seen and then expanded days/weeks later?  My agar experience so far has been puffy growth supposedly cobweb mold with no sectoring.

I think this one might be cobweb but im not sure yet. Im gonna let it grow out and see how it looks. It started out with a ring like pattern similar to a bulls eye. The strands are sort of hairy or wispy if that makes sense.


Im pretty sure that this one is mycelium. It looks similar but it looks more puffy and strands are closer together and it hasn't grown as fast or aggressive as the other one.


You probably won't have any sectoring after germination. I don't think that normally happens until after a few transfers.

Edit: I should mention that this is my first time trying agar and it will be my first time cultivating. Donโ€™t use these photos as a reference or guide to help you determine what's mold and whats mycelium until someone else with experience can verify it. For all I know, they could both me mold or both be mycelium.

Yes.

Fungi have a few different "mating types" as mycelium chains and are usually just referred to as "positive" and "negative". When hyphae from + and - meet, they combine their nuclei to change the structure from a haploid cell to a diploma. When the fungi wants to reproduce the 2 nuclei fuse together and convert the cell into a spore.

As long as its the same species, there shouldn't be a problem with producing fruit bodies and spores.

Until you get into the freaks that don't produce spores and shit of course

You can just mix different spawn into a tub thatโ€™s the easiest way.

How do cultures differ on a high nute plate than a regular "half strength" agar plate? Does the growth look different? And how so compared to regular?

Hey there, just thought I would share some pictures of my spores on agar 8 days after inoculation. First growth was spotted 3 days in. the dark spots are clumps of spores which dropped out the syringe. Pretty impressed with the spore syringe though. really clean. looks like I won't even have to transfer. What do you think?

The growth tends to be slower and more compact with more nutes. Also more likely to go tomentose.

Oh wow really, because the the recipe I used from you (7 grams agar/ 10 grams LME) already gets pretty fast growth I feel... So using the common โ€œhalf strengthโ€ ratio of 10 grams agar and 7 LME will be get even faster growth??

I have a old swab tried to put on a plate its been over 2 weeks and nothing.  Could i cut a piece of the swab  off onto agar?
