I could be wrong on this, but I think you would have to add some dextrose or another form of simple sugar. Also it is a flour so I would think the solid pieces would have to be filtered from the liquid (?) Prob be much better and easier to get a LDME (light dry malt extract)

Not sure how well you can tell from the pictures. I poured around 30 plates a few days ago. Didn't seal them and put them into brand new gallon Ziploc bags until ready to use. 1 gallon bag had 10 plates so i broke it open 1st. In my SAB, I continued to make a couple transfers, and also take a vendor LC to plate to establish the culture on agar for further expansion and saving.

noticed the other day these pink specs all over the place. even on 2 plates that didn't end up being used but wrapped up at end of the session have the specs

the other gallon bag that has the rest of the unwrapped bag doesn't appear to have the same pink stuff on it :shrug:

any ideas? yeast? or something else? I wish i had made plates w/o the coloring to get a better idea of what color the contam is for sure

Hey everyone, thanks for the advice yesterday. Once I got home I looked around for some tools or wire to bend and low and behold, this set of picks i inherited had just the right thing. It's a pointed pick, that has a bent end with a 25-30 degree bend inside that near the end. Its great for allowing me to scoop and stab the side of the wedge and then slides out perfectly when I place it onto the new plate. I just did a set of 16 transfers and it seemed to go a lot smoother and quicker.

Now if I could just stop hitting the fucking arm holes in my sab...

Weird. It almost just looks like sediment that got dyed darker with the food coloring. But that first picture kind of throws me off to see it in the streak like that.

The little red specks in the other pictures look like particulates and not contam at all...

cam I had one of those specs show up in a no pour with grain water .
I’ll post a pic later but looks identical to yours except I only have 1 speck of it and mine was at the bottom of the agar. (I pour kinda thick)
Ben watching it for like 3 weeks now it hasent grown at all.

1st I thought maybe it was from cooling the blade in the receiving plates, but like dirtybird mentioned, the appearance in the streak plate kinda blows that theory

Ill keep an eye on them and get the cultures moved on asap

I’m pretty sure it’s just sediment from the agar/nute mix. I tend to get those when I don’t shake and boil it really well before pc.

they weren't there prior to making the xfers to them
and the rest of the batch in a separate Ziploc still doesn't have signs of them on there yet

I must be looking at something different. So much weird stuff goes on with plates.

I’ve had some with lots of the sediment and little strings that were fine and some that looked like bacteria was slowly spreading out from each one in the middle of the agar.

Did the unused ones that got it get opened at all?

I am trying to start gaining some experience with agar. I'm reading like crazy but have not been able to find an answer to a quick question, absolutely newbie question...I will be using premixed MEA. Do I need to heat it order for the ingredients to mix properly or is it better to mix it with cold water, put it in the HG containers and PC it? Thanks!!

Mine comes out much clearer when I dissolve it in cold water really well and then boil it in the microwave before pcing.

I got some funky looking growth on my t1 aa plate. I transferred from a really rhizomorphic section of the germ plate, and It just looks weird to me. Cant tell if thats mold or what.

not intentionally, cant say for certain the lid never got knocked, but they were never exposed to open air as the ones who got xfers were

ill keep an eye on them, the streaked plate is by far the worse, right in my streak pattern

the others ones I'm not as concerned about but will still be taking transfers as soon as the culture grows out a tad
