Forest fires release nitrogen and promote Nitrococcus and nitrosomonas bacteria even years after the fire, all cellulose digesting fungi need nitrogen to aid in Laccase enzyme production to convert cellulose and lignin into lipids and sugars as a source of energy that’s how the nitrogen cycle works (nitrogen/carbon)

so there’s your answer

Wtf? I didn't ask a question I stated a fact. Also to get something straight with you morels are not all fire morels or dependent on fires. "Naturals" do not require fire and there are no forest fires in Missouri or the east coast of the USA.

Quote:
to why fires help morels and why Owers uses urea it’s the nitrogen, there’s even several sources that demonstrate morels will grow on rags dripped with urea every 3 days and also urea liquid culture, nitrogen is a critical aspect of wood/forest fungi

what the hell are you rambling on about? Why are you preaching to the choir?

Found some gray morels and would like to try to clone them, what are some good techniques for cloning them on agar?

You could try to take a tissue sample preferably from the inner tissue and slap it on your agar.
Only one problem tho...its not in a vegetative stage but a reproductive stage and the current state of the organism is more than one
organism due to plasmogamy. Once it is cut off from the nutrients the tissue begins to breakdown they don't go back to feeding.
Morels have an asexual stage where it can reproduce without another compatible mating type. Whether it is parthenogenic is beyond me.
And of course it has a sexual cycle...maybe. If it breeds with itself it's either asexual or parthenogenic.

Shiiiiiiiiiiiiiiiiiiiiiiit, put that sand in the jar and get sclerotia to grow.

I've done this with some store bought wild black morels. I took a section of tissue, dropped it in a 10% bleach solution for 2 minutes and transfered that to grain. It grew vegatative mycelium and produced sclerotia just fine. I've since transferred that culture to agar and that agar to more grain today.
Typical LME agar should work but it is difficult to transfer clean tissue as the inner tissue is so thin. That is why I used a 10% bleach solution in an attempt to chemical sterilize the section of cut tissue. I did this with about 10 30-40ml grain jars and transfered the clean ones to more grain before I actually made agar with them. But starting with agar would be best.

I was going to make a new thread, but figured all you fine folks around here would know. The answer is probably somewhere in this thread but alas I'll ask it again.

I recently found some nice looking importuna and put some tissue on agar to clone and hopefully cultivate. I was wondering though, since morels are Ascomycetes, the vegetative mycelium is monokaryotic, while the tertiary and mushroom producing mycelium is dikaryotic.

So, would that mean when putting a tissue sample from a mushroom, making it transition back into vegetative mycelium, two separate monokaryotic mycelia would grow?

Yeah that's the big question. How can something that is dikaryotic be vegetative when its trying to go through a reproductive cycle?
And if it's dikaryotic it would be relying on the nutrients it stored up in the sclerotia to form an ascocarp.
Otherwise why would it need to store up nutrients to form a fruit body? It obviously isn't in a vegetative form when it is an ascocarp.
So I question that anyone actually got morel mycelium from tissue culture.

How closely did you inspect your culture? I suspect that the tissue contained spores that resulted in mycelium growth.

Yes that is very well possible, I was going to reply stating that. But I would think the 10% bleach solution would kill any spores present, morel or otherwise. Perhaps the contact time wasn't long enough for a complete kill rate. But I know this is the process and percentage used when cleaning pollen sacs for use in canola tissue culture in order to sterilizing the pod before opening to extract the pollen and plate it on. At any rate putting spores on agar would work too.

I plan on getting back to some Morel experiments this year. Like I'm sure many here have, I've cultivated sclerotia from both Yellow and Black Morel strains and have had very limited success with outdoor transplantation techniques using spore slurries, stem butts, and spawn. Because the Yellow and Half free Morels are now popping in my area I decided to kick it off with a brief experiment. I dug up 2 very small Yellow Morels including about a cubic foot or so of surrounding soil and transplanted them, their soil, and attach sclerotia into pots and put them in a fruiting tub at 65 F. One was left in open air, the other I covered with a clear plastic cup to limit air exchange. Light was 12 on 12 off. I wanted to see if the mushrooms would continue to mature if they were separated from the parent mycelium or sense that they had been "picked" and abort. While I did observe color changes indicating maturation over a 3 day period, they showed no growth and aborted. The stems rotted and collapsed. I confirmed the sclerotia were still attached and buried them in my compost pile. It seems to me they sensed the separation from the parent mycelium or sensed the disturbance and quickly aborted

I recently read some Australian research on indoor Morel fruiting that failed to induce sclerotia to fruit of several strains using parameters similar to what Ower described. They did say however that primordia formed on two PDA petri plates after being refrigerated for 11 months at 4 C. Interesting...

Here is my current thought process, and what I want to be the focus of my research this year.
1. Select a saprobic species, importuna, rufobrunnea, etc.
2. Select an aggressive strain from conducive species that aggressively form sclerotia
3. Cultivate sclerotia to maturity so that they are nutritionally capable of supporting fruiting.
4. Attempt various methods to induce carpogenic germination of sclerotia via external nutrient sources and other potential stimuli (I have several whacky ideas of how to anger my sclerotia)

Im interested to know what others thought are on this topic though. Do you think that wild morel sclerotia are "programmed" to fruit or not based on conditions during their formation? Or do you think it is the conditions present during the fruiting season that signal them to form fruit or just germinate more mycelium?

I hope that the pictures are not in the right order ...

The fruiting body of the fungus is like the apple of an apple tree ... only the fruit. If you pick up an apple out of a tree, take the whole tree to your garden, plant the tree and hang the apple on a branch ... can it keep growing?

Before speculations arise ... that was a rhetorical question ... the answer is NO! ... and because of this the morel couldn't keep growing. You cut the morel off his mycelium (the tree).

No need to simplify the life cycle basics of higher fungi for me. I'm not a professional mycologist but I play one on TV. Pics are out of order yes, only supplemental to what I wrote. Fungal life cycles are actually much more complex in many instances. With oyster and shiitake log totems, you can separate the log chunks once mushrooms pin, essentially breaking up the parent mycelial colony, and mushrooms will still grow and mature. There isnt alot of data Ive seen out there on how sclerotia behave if separated from the parent mycelium. My little experiment was simple because the mushrooms were available and the results were not at all unexpected. My guess is many terrestrial mushrooms would similarly abort due to disturbance, break in mycchorizal relationship, separation from parent colony etc. One reason I wanted to tinker is because Ower's process calls for the harvesting of mature sclerotia before induction, essentially separating the sclerotia from the parent colony before inducing mushroom growth, I was interested to see if the sclerotia would continue to support fruit development when removed from the parent colony. This also applies to the question if morel sclerotia are true or pseudo-sclerotia.

Does anyone know if Pleurotus tuber regium, or other similar species produce fruit from harvested sclerotia?

That would be a great solution if the fruitbodies were persistent all year round but sadly that's not the case. The natural state of the fungus is a mycelial stage feeding and storing nutrients up underground.

I will agree with this. That seems to be what happened with the one in my signature photos.
Only after eating all possible nutrients in the PDA.
