Just did my first transfer, was more tricky then I expected.

People lifting 5-6 dishes and starting from bottom. It was extremely hard to pick up that much, was slippy AF.

To take agar with mycelium also was pain in ass to pick/cut in pieces :laugh:

Whole procedure look so not clean... we will see, did 16 transfers from 5(spore agar)

Having some issues with water pooling on the surface of my agar with pasty plates.  Following the tek to a t.  I was going to do a few transfers today but realized the plates I made a few days ago are most likely unusable.  I opened one to get a clear picture of the water, I'm sure it would hinder myc growth. 

I'll most likely dump this set and make another tonight, just not sure what is causing this much water on the surface of the agar.  Should I remove them from pc, unwrap foil and remove paper towel once cool enough or let them cool in the pc overnight?  Any pointers would be appreciated.

Condensation is generally a function of temperatures.
Temperature poured dictates the amount of vapour in the dish
Temperature stored dictates how much condenses on the surfaces.

I'm told that condensate doesn't impact growth and hasn't when I have experienced.

Do you store your plates upside-down? In your SAB you can crack the lid and drain. . .

What causes these rings in the mycelium? Is it temperature fluctuations? I thought these plates were looking okay but the only other time I've seen the rings is in mold like the picture on the right, now I'm worried I'm growing mold



Also is this just the mycelium eating the food colouring? Started as a green wedge.

These are shiitake, king stropharia and oyster plates not cubes

That plate in the 2nd pic is growing death.

Idk why them rings happen. I get them now and then. Looks pretty severe on them plates in the first pic.

My plates dnt see a lot of light. I heard its from light cycle. Might be from temp.

Acremonium grows like that. Im wondering if a culture contaminated with it will grow like that.

Message them, Amazon will probably send replacements. This is almost guaranteed to be true if it was fulfilled by Amazon and not sent directly by a third party seller.

it worked for us! ordered celltreat brand, seller was pipette, one sleeve had a hole in sterile bag and one sleeve had a cracked plate. sent a quick msg to seller about it, expecting the unsterile sleeve to be replaced, they contacted celltreat which sent two full sleeves to replace, very well packed and arrived in 3 days from msg to seller.

very pleased with that transaction and used the sleeve of unsterile plates to practice pouring.

This is the first transfer I did from a TOC MS syringe. The regions of growth marked with stars are due to me rubbing the transferred piece of agar on the plate trying to get it off of the scalpel.

I'd love to hear your thoughts about
  • The fuzzy growth growing straight up off of the transferred pieces (most prominent in colonies 1 and 2)
  • Where to take another transfer from. I'm thinking that colony 3 looks the best; I like that thicker growth (ignoring the starred regions)
  • Anything you think is important for me to notice!

Anyone know where nutritional starvation begins to become noticeable?  I ran a bunch of dishes with 1% LME against 1.28% LME and culture growth was the same.

3 looks like it has bacteria or some weird mold myc riding along, maayybeee. I wouldn’t use it since you have 2 other worthy samples.
I would transfer from the 9-10oclock position on 2, and if you have extra plates the 9 and 3 o’clock position on 1 just to see what’s up. And I’d do it quick so neither of them end up touching sample 3. I could Ben paranoid about it but it looks iffy to me.

I think two is the best bet, the others look like mold is hitching a ride.

Sometimes the first transfer looks sketchy, but to me it looks like something is meshed in and growing way faster. Don't be afraid to start over, I would make a new batch of plates, transfer from 2 and go back to Ms on a few.

When you empty that syringe, make sure you save it. It comes in handy when you have sketchy plates. Look up josex biopsy Tek.
