Difficult to tell with light behind it. Must have pic of light on it. Looks kind like acremonium with the thin cottony myc and them rings, but sometimes cube mycelium makes them rings too.

Got a couple more with light from above, hopefully this helps. these are from some B+ spore prints i made about 2 years ago and surely are dirty but my last syringe of ecuador is producing nothing on agar (not even mold) even with different streak patterns and recipes of agar. I might just need to accept defeat and start fresh with better spore prints.

Oh, i forgot wats up with that. The myc travels under the surface or something. Almost looks like when spores start popping off before the mycelium pops up. I wud give it a few days and see what happens.

Sorry Gypsy, meant that for pureshrooming

Two years isn't very old but I'm working with prints from 2012 and the original plates looked a lot like that. Couple transfers to different nutrients on agar and no change, then I used it to inoculate wbs jars and the Mycelium appears rejuvenated.

First agar was snake shit and beer. Second agar was Karo and beer. The jars I spawned to were made with 1000ml dry wbs/500ml coffee.

Wtf is going on here? Feeling cantankerous this morn.

And it makes me nervous to be hearing folks go on about grain soak. Grain soak agar is bullshit for anything other than identifying a section whose mycelium looks better than the rest of the culture. Mycelium cannot be trained. If mea isn't doing it for ya, pda prolly will.

Well, I'm new to this, but grain soak is listed at the #1 spot in Bod's Comprehensive Agar TEK so I guess it works for him. :shrug:

We were talking about the last pic I posted on this thread (link). It's MEA vs oat water (from boiling not soaking) agar. MEA is red. Both transferred at the same time from the same dish, and the same pattern holds for all other 12 oat & 28 MEA dishes I made that day. So you think the myc is just not liking it (too much/little nutrition/fungicide presence/etc.) instead of needing time to adapt to a new food source?

I dnt knw exactly what is up with the growth on them grain soak plates but that's what im talking about. It's gonna look all fucked up. If you lower the nutes way down you may be able to get a few sections on a few plates to look normal. As always, do as you wish my friend, but it is my suggestion that you stick with mea or pda.

These plates are 10g agar 10g LME 500ml water

Where do you think I should grab the transfer from? Leading edge or just anywhere?

Appreciate the replies! I've done really well with MS syringes up to this point but I'm really hoping to get agar down and change my game

pureshrooming, I'm currently waiting for my first jars to colonize, but FWIW, 20 days ago I also started from a B+ print, using the exact same agar recipe as yours, and had a similar pattern of undifferentiated, fully submerged myc on all 16 dishes I made from the print. After 1-2 transfers I started getting more familiar looking cultures (like I see posted here), so I'd just go ahead and transfer away. Whether it's contaminant removal or strain reduction, the transfers seem to do the trick.

Idk where the best place would be. You can see where I took mine. When I take transfers from unenthusiastic myc I only take a tiny amount. You can see how small the transfer was in the center of that plate, tiny. I believe both those pics are t1 and t2 in order. Both transfers grew out really uniform, no sectoring. That gouge in the agar was from starting several other plates I hardly used any.

I chose to take the transfer at a point where the Mycelium had colonized all the way to the bottom of the agar. I don't like transferring uncolonized nutes from one plate to the next this is why I don't go past the leading edge and the edge on my plate looked like it was just barely skimming the surface. You can kinda see it in those pics. I don't know if this is the best choice but it was my choice and that's why I did it this way. Maybe somebody has a better way.

Well now. Respectfully agree and disagree.

Grain soak agar is shite because you can't tell what your nutrient level is.

As for 'training' as you put it. . . Are you suggesting that the myc is excreting enzymes for every possible nute source all the time? I'm led to believe that myc reacts to the nute it encounters and taylor's the enzymes it excretes to suit?

For the initial jump to a new medium, myc is using the nute it came with (agar transfer) and its own cell structure as energy until it detects the correct enzymes to use on the new medium.

Any evidence that contradicts this, please share.
