Thanks a lot Edmunter. I understand and agree with all of that. But as far as contamination goes, the plates look clean and ready to go to grain? I’m just trying to get one or two nice fruits to clone, as quickly as possible, so I’m okay with doing a2g with all of these plates instead of spending a few more weeks doing transfers.

Thanks again!

Got some LME at a brewing store today, and will be making some plates tonight.  Usually I make my Agar from with GSA or BRF so this’ll be a first for me.

Planning on 2% for both the LME and agar - thinking of throwing in some food coloring to make spotting contams easier.  Question: what color do you all use (I see a lot of red and blue), and about how many drops for 500ml do you guys use?

I use blue, and it doesn't take much probably a couple drops, 2-4 will probably be enough. I use that gel food coloring, might be more condensed.

As said, id go one more transfer on said plates and take a chance with 1-5.

This is a clean plate

Its not like the growth id like to use for a grow yet.  Maybe someone else can chime in here.

Idk what my suggestion means to you, but that's a beautiful plate and if i had a beer that day it would probably go to 4 jars. If i were sober and thinkning correctly, only 2

I have 5 plates of Argentina variety that should be ready to transfer to grain in a couple of days (this is the third transfer from MS).

Trying to learn more about what to look for on agar. There are no bacterial colonies, so that's good...

Anything I should notice that would prevent me from going to grain?

The odd shape and resistance to grow in certain area.  If you transfer the healthiest mtc you are taking healthy genetics and losing the slow creepy ones.  If you transfer the fastest growing myc you lose the slow stuff.  If you transfer the pretty myc you lose the ugly myc ect.

Using 1 and 5 gives you room to reduce the genetics in the others.

Those fast growing bits on plate 3 at 12 oclock, 2 and 5 are what you want.  If you take the smallest amount you can you are reducing genetics.

Plate 4- 2,4 and 6

Thanks, I was trying to show our fella what a clean plate looks like.  What do you think of his plates

From your last post it seemed like the reason you wouldn’t use them for a grow yet was the genetics.

I’m still unclear on whether or not you think my plates show signs of contam.

If yes, what specifically are you seeing that suggests contam?

Your example is very rhizomorphic, but a culture doesn’t need to be rhizomorphic to be clean, as shown by the fact that you said my first and last plate look good.

If my plates have contam, I want to know exactly what about them suggests that : )

Or if the reason you wouldn’t use some of my plates for a2g is simply because of genetics, I’d like to know that too.

Thanks!

Sorry, i guess you didnt see my previous post...
Those plates look very fluffy and uneven to me. A Trusted Cultivator has told you take transfers. I agree with that. If you must, put the best looking one to grain, probably the 8/9 o clock position on plate number 3. The rest of them need to keep getting cleaned.

Edmunter, if that is fleshtone underneath you last post then it's probably fine. If the mycelium is that color, i would say no lol. I stopped using food coloring because it was throwing me off  personal problem

Sorry for the confusion, but I was replying to Edmunter. (Who is the TC you speak of?)

All I want to know is if Edmunter (or anyone else) thinks my plates might be contaminated. And if so, what exactly about them indicates they may be contaminated. People keep saying "do more transfers", but they aren't clear about whether they recommend that because I can achieve better genetics OR because there appears to be contamination on the plates. I'm asking specifically about potential contamination and what it looks like. Are you saying that "fluffy and uneven" are signs of contamination?

I'm particularly curious because GypsyCurse told me here that my plates look fine for a2g.

Thanks : )

They dnt look contaminated to me but that doesnt mean they arent. I thnk thats why we may have a difficult time getting behind a "go ahead man!"
I wud put them to grain. I see nothing that wud prevent me from doing that.

And if done correctly, transfering gives the opoortunity to spot shit in your shit, or simply get away from it.

Would give me concern about being contaminated

So with the above edit above I would use 1 and 5 for grains as they are more uniform and transfer the rest for safety.  This way you are covering your arse   What you want is uniformety and to be able to see the mycellium clearly.  If you look at my 2 cultures above 1 is clearly clean and the other im 95% sure.

You only need 1 clean jar to expand to 100s

So, most of my agar has been super wispy growth. Here is a pin clone from an earlier plate that i also posted in this thread before.  A few more pins came up and i transferred.  This one is the best looking so far and will serve as a basis for other plates that I'll do A2G.

It's interesting to see the rhizzo growth and how it branches out after transfers [done about a week ago or so prior].

Would you do more transfers?  How would you go about preserving this plate for a long time?

Sometimes that can be the mycellium running away from something.

I would definetely do more transfers.

Once you have culture you want to keep you can just put them in the fridge.  Check them  every couple of months or when you reach in for a beer and pull out  plate instead.  If they dont look as healthy tranfer to another plate.  They can last 6 months.  Longer that this and you need to slant it.  It has to be a decent culture imho to bother with it.

Still on my journey to identify healthy mycelium. Admittedly I am still not sure what I am looking for but with this thread combined with another contamination thread I believe I am looking for threads spreading outwards not just fuzz and bubbly blotches. I took a few shots of my plates from the back in hopes the powers that be on this forum can point me in the right direction.

I am going to be doing transfers until I seclude a healthy plate before dropping into grain. So here are the candidates that I though had some potential

Edit: most are spore prints onto a sterilized cotton swab except the 3rd picture. If I remember correctly.

Im srry dude, but the light behind shot makes it really difficult for me to tell wtf is growing on them. Maybe light from side or above.

I can tell you that germinating spores always appears to me at first as a slightly opake milky bump in the puck surface.

Can mycelium grow ontop of mycelium? I had some trouble making this transfer and ended up dropping a wedge. This has been steadily growing for about 2 weeks, stayed bright white the whole time.

Is there any molds that grow white and don't sporulate on agar? I have some suspect plates I've kept around that have stayed nice and bright white for months
