Li( liquid innoculant) is made using sterile water and agar, v.s LC which involves adding nutrients

Here's a link to the Tek I use.

You can skip buying a blender if you have an impact gun. You use the little square bit that ratchets attach to. They sell oster blender blade bases on Amazon for pretty cheap.

Happy Halloween! I watched The Taking of Deborah Logan last night for the first time. Pretty creepy if you've been looking for a good movie. I like those found footage style movies, like As Above So Below.

Anyways making LC for the first time in a year 😎

QQ, those of you with All Americans, know of a good replacement trivet for their big boy version. I forget what size it is. I think like 43 quart or something. Like an aluminum one?

I've done rev's before with good success. It's pretty easy too. Just cut up a plate and put it in sterile water.

Unless you have a flow hood, it's a good idea to treat them as single use.

A free-pour of Pan Cyan BVI would fully colonize a jar in less than a week.

Magnum PE in a 27qt tub. I'm letting these go for as long as possible to see how big they get. This is only 1 week from pin 27 days from spawn.

Here is an image I took just now while observing a gill fragment from Psilocybe 'subsecotioides'. I have been scoping the fruits from my bed every few days to be able to determine macroscopically when the fruitbody is most likely to have the ability to eject spores and produce an actual spore print.

'subsecotioides' is a secotioid mushroom meaning the caps do not fully open like other typical Psilocybes and their basidias can barely eject spores. Considering they lack the ability to spore print, for the most part, they are generally swabbed for spores.

However, as you can see in the picture I'm sharing, there is pronounced hyphae with clamp connections. When clamp connections are present, that means the culture is dikaryotic and has mated. From there, it has the ability to produce fruit bodies. No clamps means no fruits. These clamp connections exist in all Basidiomycota fungi, which includes the genera Psilocybe, Panaeolus, Gymnopilus, Amanita, Inocybe, Pluteus, Conocynula and many more.

The hyphae are always present during development of the fruitbody. There is no one time you can swab and have a higher likelihood of avoiding hyphae. Besides, you want to wait for the fruit to mature anyways to be certain you actually grab matured spores.

Now the problem is when you have a fruit that won't spore print, your only option is to swab if you wish to return to a multispore grow, or want to obtain a monokaryon for breeding. However, when you swab, you have a very good chance of grabbing hyphae and actually cloning said mushroom when you go to streak a plate.

Your only true course of action at this point is going by date and hoping for the best. This is why you've probably heard it mentioned on here several times before about the argument if the growth on the plate is a clone or if it's truly multispore.

Growth in just a few days is probably a clone.

Growth in a week or more is probably multispore.

It's only a "probably" because no one can actually say with certainty. This is just one reason why you should always date your plates.
