I noticed something very interesting just now! I cooked some LB Tek trays in my pressure cooker nearly a year ago. Well I had to move the PC maybe a month ago to fix an outlet behind my range. It was heavy so of course I looked inside. All of the trays in there were perfectly clean and healthy looking. Amazing right!? Well I just went to take them out just now and now they are contaminated!

What I think the reason is behind it is that the PC does not pull nearly as much vacuum as we suspect it does when cooling naturally. The inside of the PC remained completely sterile after all of these many, many months.

Now, when I opened it about a month ago, the inside became contaminated. The filter material I use on my LB trays are not hydrophobic. Considering they were wet, they were able to draw in contamination at that point.

I thought it was very insightful. Use that information how you will 😎

Went out an hour before sunrise today to hunt for gyms with TRUE uv 365nm flashlight. It's really fun doing that because gyms glow.

No pictures unfortunately because the shroomery isn't letting me upload again :frown:

Here is an image I took just now while observing a gill fragment from Psilocybe 'subsecotioides'. I have been scoping the fruits from my bed every few days to be able to determine macroscopically when the fruitbody is most likely to have the ability to eject spores and produce an actual spore print.

'subsecotioides' is a secotioid mushroom meaning the caps do not fully open like other typical Psilocybes and their basidias can barely eject spores. Considering they lack the ability to spore print, for the most part, they are generally swabbed for spores.

However, as you can see in the picture I'm sharing, there is pronounced hyphae with clamp connections. When clamp connections are present, that means the culture is dikaryotic and has mated. From there, it has the ability to produce fruit bodies. No clamps means no fruits. These clamp connections exist in all Basidiomycota fungi, which includes the genera Psilocybe, Panaeolus, Gymnopilus, Amanita, Inocybe, Pluteus, Conocynula and many more.

The hyphae are always present during development of the fruitbody. There is no one time you can swab and have a higher likelihood of avoiding hyphae. Besides, you want to wait for the fruit to mature anyways to be certain you actually grab matured spores.

Now the problem is when you have a fruit that won't spore print, your only option is to swab if you wish to return to a multispore grow, or want to obtain a monokaryon for breeding. However, when you swab, you have a very good chance of grabbing hyphae and actually cloning said mushroom when you go to streak a plate.

Your only true course of action at this point is going by date and hoping for the best. This is why you've probably heard it mentioned on here several times before about the argument if the growth on the plate is a clone or if it's truly multispore.

Growth in just a few days is probably a clone.

Growth in a week or more is probably multispore.

It's only a "probably" because no one can actually say with certainty. This is just one reason why you should always date your plates.
