I don’t think it matters. There are also hyphae below the surface of the agar you’re transferring from and beyond the white edge, so as long as you take small samples, it will be fine. And even if your sample is too big, it’s not a problem either because it will just take a bit more time to get to the agar on the target plate.

My transfers always end up on their side I think :shrug: my last cut is to scoop the transfer out (so it’s kind of flat) and then I stick it in the target surface vertically to get the transfer of my scalpel.

I’ve done side by sides of mycelium up and down and in the end it didn’t make a difference.

Leucistic Eungella Australia plin from a clone plate that I used for g2g and LI. Took a few samplers first.



Now, this was a clone already. When you clone a clone, are you actually changing anything meaningfully, or not really?

This Charlie Brown plate is pinning hard. A bit unorganized looking. Sketchy or fine?



These last two batches of agar have been iffier. I had done one premade MEA mix that I had laying around, and I really liked how rhizo it was. But I hated how soft it was (and having to buy ingredients that I don't otherwise need).

I'm back with my home recipes, using Golden Coins PURE Agar Agar,  but these last two batches are getting less rhizo growth than usual. Could be a coincidence, maybe Golden Coins is not ideal (they don't have telephone brand anymore at my local store). I have noticed that things get wet when transferred, not just from condensation for new plates, but maybe permeability of the agar?

Anyhow, I will do a pasty plate recipe for pours next and see what happens.

Dude I must have owned 5 French presses over the years but was French Press-less when I read the tek. So I bought one, again. And goddamn I'm wondering where the other ones went and why I didn't have one because they are great for lots of things (I don't use it for coffee though, haha).

I guess there is nothing wrong with the recipe I am using it just takes forever for growth directly from LC to agar as it seems all the LC needs to dry out or the myc eats it before the mycelium start moving on the plate. Doing transfers to the PDA plate take off in 1 -2 days. I have noticed I poured some plates a lil thick as transfers are almost touching the lid.

Not sure if penicillium vs trichoderma in the middle area? Definitely a shade of green.

Would this be a good situation to practice making clean transfers away from the contamination? Or just trash and start over? Sorry for blurry-ish photo.

If you have genetics to start over I would, probably sporulated and mold is hard to clean up.

Soo unsure if the off color on one of those circles is metabolites on this plate or if it's slightly bacterial πŸ˜… what say the pros.

It's psilocybe caerulescens if that helps

Looks like there is some bacteria in there as well. Also what are these faint trails/fine lines visible on the left of the plate? Could it be you are having a mite infestation or is this just an optical illusion?

TBH the white to the right doesn't look very promising either. I'm with Kloakk and would not recommend to try cleaning this up.

Was this a syringe inoculation?

The lines on the left are on the plastic lid. Could be my finger nail creating a line on the plastic lid. I smushed my finger over the lid and it disappeared and created some new ones with finger nail.

Also, inside of lid could have some marks from from condensation that has dried up (maybe also hard water from the PC, I saw I could use some vinegar to help with hard water). I also tried adding some carbon to the agar, but that didn't work too well because all the carbon separated to the bottom after cooling from PC.

Yes, this was from a syringe inoc. I recently learned the type of SAB I am using is very bad. Pretty sure I contaminated the agar in SAB. It's the flimsy soft plastic that can move.

So, went to the store today to get a nice rigid plastic tote for next agar attempts. Thanks for all the replies!

Yeah, all good in the hood and good to have a nosy on the org and seeing you guys doing some cool shit. I just expanded two pan LCs from my culture bank. One 5.5 Thai Suphan Buri and one 2.5 year old Bisporus. Hoping these are going to perk up and I can do a few tubs and also doing a little culture work with Hericium which I finally managed to get from a wild specimen onto agar without being riddled with mold. Might have to change the agar recipe and put a little more than 0.5% LME in, it's slow and thin. Well, off to the gourmet section to read some....

Glad to hear things are good man. No more pans for me this year, but I did manage to grow and print Bisporus, Chlorocystis, Weza and komodoXBVI this season so I ain't complaining even though Mystery SA gave me nothing but trouble and headache :lol:

Oh and can't believe you had me googling Hericium just to find it's Lion's mane :rofl:

Samples of woodchips.

There's like 3 chips in there though they originated from same spot practically.

They got scattered abit on agar during transfer idk lol there's some clean my it looks like for me to work with at least on other parts of the plate that's not green :dealwithwhat:

To naked eye it has blue staining n looks white. With light under it it looks green and with a light in front n looking at back of it it's orange.

Got it, well if it's from the same patch then just take what's looking cleanest and do a transfer asap incase there is something else brewing on the plate :rockon:
