Does anyone still sterilize there tools in a PC anymore?

Like scalpels wrapped in foil? Or is alcohol & flame sterilization good enough?

ill sterilize qtips and certain tools ill want clean like loops to dip in lc for testing but i havent pc'd a scalpel in ages.

Cubensis mycelium bridging the  3mm gap of an agar trench, on MEPA. In the background of the trench is the black metal of scope stand/base. 2025


A dried colonized whole oat grain inside of an agar "cabin", on MEA, rehydrating, about 48 hours after placing onto agar. 2025 [Thanks for sharing your dry grain storage method, Fahtster!]

Stunning pics flapping_bird :super:

Have you heard of storing bacterial plates vertically?  The myc can fight gravity and climb upwards, but the bacteria can't.

Thanks! They were fun pictures to be able to capture. ***ESPECIALLY*** that semp clone pic, with thanks to you! I feel like I’ve won the lottery on that one, just getting to the point of being able to take the sample to put to agar. Excellent resources, thank you! That’s a golden tip.  I haven’t used PDA much (only this current batch of plates) and surely it can’t be anything specific to PDA, but I have noticed on a few similarly dirty clone samples on PDA that the myc eventually seems to gain the ability to outgrow and ?kill?/?encapsulate? / ?overgrow? the baccy spot and continue on it merry way, looking pretty good in spite of it.

Can i put 1 grain from a grain jar im putting into spawn on agar and grow that out? Just do it carefully in front of a flow hood? I know you can grain to grain but I don't know yet If i want another jar going quite yet. But I also want to have it on hand ready to push to grain if I wanted to.

I just got some 500ml media bottles and I'm gonna make agar in them for the first time. When I take them out of the PC, is it safe to tighten the lids down then?

I thought I've read that will create a vaccum. I plan to use them as soon as they're cool enough to pour but might want to make some ahead of time for the future.

I'm probably overthinking this but I'm used to cooking agar in containers that have filtered lids.

You can finger tighten them to move into your work space, then back the lid off again. The threads act as a barrier to contamination. Don't tighten them while piping hot or your agar will begin to boil.

Thanks man, I plan to send these to several WBS or oat jars very soon! Need to learn much more about agar... I barely know anything about it lol. Just squirted some MS onto these plates and seem to be doing OK. LOVE this hobby! Just wish work didn't consume our time as I would love to devote all my time to this awesomeness! Not enough hours in the day lol

Could anyone explain what’s happening to my ochra transfers from spores?

2% agar and 2% LME plates.

I was told regarding it growing up that it was too much nutrients but what is happening with the juice?

Possible mycogone perniciosa.

Orange or yellow liquid are enzymes produced by the myc known as metabolites.

Typically excreted in microscopic amounts for exploration of surroundings, digesting food, and in this case, dealing with stress, likely from contamination
