Inoculating agar with a liquid culture?
    #4953905 -

Will this cause problems?  Or should you use a colonized grain kernel?  Trying to figure if it would be better to inject a colonized quart jar with sterile water, shake it about, and then draw the water back up into a few sterilized syringes and then just inoculate some spawnbags, then spawn the grain to pasteurized wood (this is for black poplar).

But I'd sort of like to play around with some agar, as well.  So...can I use this sort of liquid culture to inoculate agar, or should I sterilize some tweezers and pick a grain or two out of the jar and use that for the agar inoculation?

    Ta, luvs.

agar can be inoculated with just about any mycelium culture.

Just didn't want to flood the plate...sometimes the mycelium in the syringe is a bit chunky...so you can pump out say, 5ml, and get very little mycelium. That's the worry--putting too much water onto an agar plate in the process of getting out some mycelium.

Shaked your LC before inoculating. A liquid culture in a jar can be agitated with something in the water (marble etc) or a magnetic stirr plate. EDIT this will more evenly distribute your mycelium chunks. If done correctly, .25 CC sill suffice

Syringe; after filling your syringe, place an alcohol swab/paper towel soaked in alcohol over the needle and pull in a half cc of air.
Careful not to saturate towel, pulling in alcohol can/will kill your mycelium thus hendering your efforts.

Cheers.  I'm working on just using magnetic stir bars in my jars from here on out.  This one's a bit different, tho, as I'll just have to shake the hell out of the syringe as it's coming from a colonized grain jar.

No worries...I've got four of them going now.  Hopefully I wont fuck it up 100% of the time.

If you have STERILE SWABS ?

Simply drop a drop of the culture solution on the swab, then streak the swab LIGHTLY across the agar plate.

This works BETTER, if you have an LC that was shredded by a stir plate stir bar. As the myc is chopped into minuscule pieces.

If you don't have sterile swabs, simply push the syringe plunger down, a teeny weeny bit, until a tiny drop of LC forms on the needle.

Then use the needle to drag the drop across the face of the plate.

If all else fails, drop a drop of LC on the center of the plate, then tilt the plate & try to roll the droplet across the face of it.

As I understand it, you injected sterile water into a (partially filled) colonized wbs/grain jar.

I would suggest you quickly use sterile syringes to extract all the solution possible. Then refrigerate those syringes.

All that water, in all that grain - is going to cause gooey problems, if left alone.

Thanks, man.

I know any colonized jar injected with water is to be forgotten.

I'll give it a go.  I'm going to wait until there's very thick colonization, then inject, say, 200--300ml, shake it about in the glovebox, then pull out a few LC culture syringes from that.  Ditch the jar afterwards.  I'll try to spawn the other fully colonized jars to pasteurized wood substrate.

Also: I FUCKING MANAGED to clone a Dryad's Saddle into LC from the stump out back.  I'm a bit excited about this--first time.  :smirk:

But that shit is colonizing the spawnbag fine (one out of like 6 jars of LC dryad saddle attempts).  So I know which jar is active.  I'll play around with that one too.

I'd just like to get some experience with agar cultures.

Thanks, tho.  You're a fucking bastion mycoknowledge.  Hats off, sir.

Also...that pic sort of looks like a butt.   That's the OTD coming out in me now, tho.
