I flame sterilize, you can see @0:02 in the video very clearly how far down the handle I go with the torch - I start at the bottom and get the tip red hot.

I don't use much IPA and have a FFU to keep the air moving. I started to wear a mask too if my partner is around because they'll sometimes start talking to me and I don't want that to affect my transfers.

I honestly do not understand why people have a hard time using a scalpel to cut and transfer agar.  It's a really simple process, just plan your moves and execute them smoothly and slowly. :shrug:  I have ptsd shakey hands and can do it.

I agree for a single transfer using a scalpel is easy imo but doing many becomes a pain especially if the wedges are larger.

Genuinely love the video, buddy. I thought about asking if you minded if I linked to it (or made a short gif from it) to use in my LI tek. Nicely done.

I mostly use scalpels with a #7 or #12 blades, but find the different pimple poppers really convenient for various things and will often have all the options handy.

I just started leaving the oldest center area from the transfer on the plate for LIs and grain inoculations. But I don't know that it really makes a difference other than increasing my confidence a little.

I saw a post a few months back where somebody was using some kind of tool that cut perfectly  even circles into agar at the push of a button! Does anyone know the name of that tool? As I do more and more transfers, it is nice to get specialized tools to speed things up! :takingnotes:

It's an agar punch. I have one that isn't spring loaded - just a circular punch and then I pluck it with a scalpel and transfer. I dig it...

Read this quote here but "peace of mind is priceless.":D it makes sense to me too and i actually told myself before bed to do this (working around the t-fer piece) when knocking up jars today and totally forgot until now :facepalm3:, a touch upset but the plate looked so good i still feel okay:rofl:

Btw i miss your old sig. It had a huge impact on me when i started as well as your first journal, just had to mention that :grin:

Gotcha, cheers! How big are they? Do they fit in a HY-800 Bactericinerator? My scalpels fit perfectly, but I'm afraid the agar punches wouldn't if they are too big and relatively "bulky".. :strokebeard:

Here is what I use for sterilization of tools (HY-800)


It was probably the best investment next to the Flow Hood I ever made in the hobby. :grin:

Cybesa you can also look on Amazon for a leather punch set. For $16 I recently got a set that had twelve metal punches from sizes 1mm to 10mm. They work great. They had anodized aluminum agar punches on there for I think $18 each. They basically do the exact same thing.

I tried leather punches, but I felt like if I got it hot enough to sterilize, it might burn through the plate and the table before it slowed down. Too much mass. Or maybe it didn't need to be all the way glowing red.

Agar gone crazy!

I dropped this T1 plate of P Mexicana Galindoi a few days ago and I was upset because I thought I ruined it when the two transfers I put in there became loose and rolled around in the plate. I put two in the plate because I wanted to see what would happen if I put one upside down and the right side up. Looked and the plate today and I guess wherever the transfers touched, I now have mycelium growing. Pretty cool!




This first inoculation plate is Mazatapec. This one has gone batshit crazy on me on black agar, but looks super cool and a bit angry. I’m going to let the rhizo strands grow out a bit more and take T1 transfers from those.




Also, could someone please tell me what is being referred to as SSLC and LI?

No they don’t need to glow red. They are thicker and heavy and the handles will get hot and retain heat, making them too hot to handle. Ask me how I know this. :rolleyes:
You just need to flame it a bit with a cigar torch lighter. What I’ve been doing is wiping it first with alcohol and then torching it for like 5-10 seconds. Then I dip the tip in a spare agar dish until it stops sizzling.

SSLC - Semi Solid Liquid Culture
LI - Liquid innoculant

LC is a nutrient rich broth that's been colonized by myc - good for mass production of spawn and fast colonization. All or nothing though, as far as contamination, so tread carefully.

LI is different bc it's myc suspended in distilled water instead of nutritious broth. Ive never done it, but I think there's more than one way to skin that cat, as far as making it.

I thought the LC was liquid culture. But wasn’t sure what the SS was. LI I wasn’t familiar with, but can’t you store myc covered agar in sterilized distilled water?

Yep ya can. Basically just take an agar transfer to sterilized water, and you've got long term storage. Thanks for the reminder - I need to order cryovials!

Someone answered but I wouldn't mess with it until you master getting perfectly clean cultures from spore to shroom, and they're not needed unless you're doing a commercial grow.
