Man, I feel that, I wish I could do that, but I'm in a place with -20 Celsius 4 months of the year. So no can do. So when you do a big bed, do one for me.

Learning lesson & noob mistake!!!!

I poured 52 new black & blue plates yesterday. I learned not to put 500ml of liquid agar in your 500ml media bottles and then put them in the PC. Boy did I have a big mess! I probably could have poured well over 60 plates, maybe 70. My media bottles all boiled over inside creating a real mess and a real PITA to clean up. I’m thinking I should stay in the 350-400ml range next time. First time I did around 350ml per bottle and had no issues. Figured I’d do a double batch of 2000ml because I have 4 bottles. That no worky! On a positive note, the plates came out great though!

Yeah, 400 ml per 500 ml bottle is the trick. You may be able to get away with a little more, but at least for me, 400 ml is enough for 20 of the 90 mm plates or 40 of the 60 mm plates, so it's actually a pretty convenient amount both for calculations (multiple of 100) and for pouring (multiple of typical petri dish sleeve size or the number of autoclavable ones that fit in a jar).

I'm assuming your 52 plates were from two bottles, not all four - otherwise those must have been really deep pours!

400ml sounds like a better bet! They were 90mm plates, and yes all four. I pour about half the depth of the plate, which is probably too much. This is only my second time pouring plates. Next time I'll pour about 1/3 the height, or should I just cover the bottom with a thin layer? which I guess would be about 1/4 the height.

There may be some confusion by your use of “oven tek.” That usually means working in front of a warm oven instead of in an SAB or flow hood. A very unsatisfactory method of sterile work. I’m hoping you are describing something else when you say oven tek. Perhaps dry heat sterilizing your glass plates in the oven to pour agar into later?

Yes indeed I meant dry heat sterilizing. I encountered unusual influx of alcohol in my body and got confused. I drink so rarely that even a beer or two is quite disorienting.

Anyways. I'm using a SAB and last single use petris I poured turned out to be fine, so I might try pouring with glass ones also. Still quite clumsy at everything agar but slowly getting better.

1/3 to 1/2 I think is about what I do. The media pretty much comes up to the level of the lid rim when it's on...which is no help to you while pouring :lol: but a good reference to check after a pour.

There's nothing wrong with thicker agar, and all the ingredients are cheap. Figure out what works best for you and don't sweat it. :smile:
