I ran the full gauntlet of recovery efforts on the ones I had trouble with. I believe I tried hydrating for 24 & 48 hours and for 1 week without success. But they were mailed to me during very high temperatures so that may be the culprit. After those failed, I just moved on instead of testing the limits. That is why I want to give these ones that I know haven't been exposed to high temps another shot.

I have tested agar baggies at local high temperatures and even fresh hydrated myc died in 75% of the samples tested 110f(44c) afternoon highs for 2 or three days. It may just be that mycelium can't take the heat here in summer. I am getting a little tired of it my own self. 82f(28c) here today for a new record.

Could also try a softer agar to encourage stubborn seed to pop as it will help with hydration. 

Yeah, I keep mine in a small zip lock in an insulated lunch box in my culture fridge to help prolong shelf life.  They’ll need rejuvenated from time to time like all cultures but you’re always making it, it’s small, etc…just easy imho. 

Those in the pic are 118 which I believe I originally received as dry grain as well, so there’s that…

They need to be cut off from drying at some point.. when I have them just sitting in a jar with large open filters, they tend to die ~12-18 months.  When I put them in a sealed container, they have lasted 2+ years.. I put some PE and semps grains into cryo tubes in April 2022 after drying in the jar for a month or so. I will put them to agar next time I start up the ffu. Looking up those dates I discovered that the PE was the culture that produced my largest blob ever in a Faht-loc bag. I would sure love to do that again.

It would be really useful for dehydration and storage methods vs potency testing.




Over 2.5 year old PE on dried wheat grains starting to recover after 6 days on agar. Pretty stoked to see that this is an option that does work.

Noice.  It really is pretty great.  All my masters fit in a qt ziplock in swab packs with like 15ish grains per.  Every time I take a clone, I clone to 4oz grain jars to double check cleanliness before g2g’ing that to qts and when I do that, I leave like a 1/4 tsp of grains in the 4oz jar.  I have adjustable ge lids for the 4oz jars that I can expose most of the lid area and put that jar with a little bit of grains in front of a fan and they’re dry in like 3-4 days.  So every clone I take immediately has been backed up on dry grains and in a swab pack before the qts are even done colonizing.  If the clone ends up shit, I just toss the swab packs



It’s a pretty great system

This is how I make the adjustable lids if anyone is curious.. I use 3 layers of tyvek.. 2 postal top and bottom and then another layer of the kite makers fabric kind in between those.. screw on with a band lid.  When I want to colonize, I just wrap the top of the lid with cling wrap and poke a small ge hole in the cling.. when it’s done colonizing and I want to dry it out, I just remove the cling. 

Disregard the SHIP.. that was for before I had the FFU.  Now I make em without it

Final update on those dried grains. After 2.5+ years, they all grew out to this in 15 days. Already have transfers of the best looking spots.

Damn good growth! What what recipe are you using? I been having pretty shit growth using 10g agar 7g lme 1g nutritional yeast per 500ml water. Pc'd for 40 minutes @ 18psi using holy grail plates. I'm assuming you are using Faht's drying jars to dry your grain?

I use between 0.5% to 1% lme and 2% agar. I started for pans and didn't like using two different recipes. The low nutes don't seem to be a problem for anything so far.

I wasn't as methodical as Faht. I just left a few grains in the jar after spawning and put the regular modified lid back on. Then left them on a shelf for a few weeks or a couple of months before putting them in dry cryo tubes. I will take some more care going forward, but at the time I was just playing around with the idea that someone here had mentioned with no clue if it would work or not long term.

I have been successful at reviving samples stored in distilled water-5ml cryo vials for over 18 months.
