Yes and no. I’m gonna give my opinion but it may be contentious.

Last few months I’ve sent a lot of multispore T1s to bulk and pretty much all of them have given me weird and undesirable growth. T2 - T3 has been a lot more “together.” I think at least T2 is the smart option because it weeds out the less stable genes, the stuff you wouldn’t select for a transfer anyway. Taking it out a couple of transfers isn’t always for insuring cleanliness, but to also make sure your genes are a little more compatible with each other.

I haven’t read this, I haven’t confirmed this, this has just been my observation.

I wouldn't trust a T1 if I didn't have a T2 to prove that at least the sector I took from was growing clean.

Making clean grain spawn is my least favorite and most laborious part of this hobby so I would rather lose a plate than a jar/bag.

both agreed, at the same time i do think that it really depends,
t1 CAN be perfectly fine and clean.
not to give out a recommandation by any means, but well i have inoculated grain with wedges from a germination plate and it was fine, but i would agree 100%, t2 is the safe route and allows for a somewhat more stable culture.

I've noticed this in the past, but never really thought much about it. Some cultures appear to "blue" when they run out of room to colonize. Is this a known characteristic of cubensis?

This sample has room to grow, but is competing with it's brother in the middle of the agar dish.

A crappy picture of my Bluey Vuitton culture. I have to work out of a SAB rn so I couldn't get it to focus at all well.

When I have encountered that in the past, it was a penicillium contamination.

I'll use them without concern.  I know that my transfer process is clean.  If you are confident that your transfers are reliable, I wouldn't hesitate.

If the original plate was gross, I'd do a T2.

I can't find the pic.  It was a well organized culture on the plate, and the center turned blue grey.  The edges looked like good myc.

I'm not too sure if it's a characteristic of cubes, but I would like to know as well. Chiming in because I have a culture of Storm Trooper on an LMEA plate that is doing this. The rhizomorphic growth towards the edge of the plate has a lot of bluing on some of its ropes. It even pinned a bit, & the tips of some of the pins got really dark blueish. Will post a photo in the morning for reference

I think that plate was a more wet agar mix. Possibly I did 7g agar with 10g LME
I've been doing most of my plates recently in that ratio, or 7g agar to 7g LME for more wet, & lower nutrient plates

Hello! I have some fluffy (but I think clean) mycelium on T2 plates. I followed HazyHorse’s no pour tek and used D3s recipe. I wonder if, given my tomentose growth, I made the plates too nutrient dense—though I weighed all my  agar ingredients with a scale. Anyhow, I was wondering what you all thought about these plates. :tongue2: If they keep growing out nicely, would you transfer again or go to grain? Thank you!!
