yeah I have long been wanting to see a thread with some sort of visual comparison of the same culture on various agar mixes. I see here locally the difference in morphology of the mycelia can be quite notable dependent on the mix. I have developed a fondness for wheat malt extract with a bit of yeast in recent times but my sterile work is pretty rusty at the minute sadly. Must take some time to re-dedicate myself

So at the minute I have been working up this Semperviva which has been a real bitch. Here is the latest iteration, a T3



I have returned to the old paper print and started again many times, going back to like november. I threw out a lot of grain jars that start from plates fairly like this and look good in the jars until a shake or two, and then some sort of sweaty socks/armpit smelling white mold/yeast takes over. I got so frustrated with it I even spawned a jar of it just to make sure I wasn't wrongly judging my jars, promptly molded also and grain did not jump off or recover on substrate

That may be the cleanest looking plate I produced to date...

What does anyone running Semps regularly think about the quality of that plate please?

It looks good to me in person although it looks a little thin or something? :shrug: I rather not waste any more resources on this if it is garbage and try and source some fresh spores or something

My other work on other cubes, cyan and bispo looks acceptable, but I just can't get these Semps right:facepalm:

this looks as good as can be! semperviva is whispy, kind of.
whats all the black crud in your agar though??

Have not tried water agar on those Soloist, have never been much of a fan typically but perhaps I should use it as part of the toolkit more often... isn't it really only effective against bacteria and stuff that runs fast on sugar media? I mean if mold is running alongside the culture somehow I assume that carries over onto water agar still?

yes that is burnt scalpel flecks, as I mentioned, a little rusty on technique currently Chris

I am gonna try again with this one if it keeps looking like this Supa

Will update in the Semp thread when the time comes, one way or the other

Thank you fellas, it is rare I would ask someone to look at a plate, but this one has been a frustrating journey

I'd probably grab a little feather from three or four bits in that 4-6 range and then one or two of the weird stuff

The 4-6 stuff only because it looks uniform and like it is going to grow out like we all like it to mostly

the other weird myc I would have only out of curiosity to see what happens on the next plate

7 oclock.  It's the most uniform growth that has filled in it's space and doesn't seem to be meeting any invisible obstacles.

Hoping it will be as clean on the grain as it looks to me on the agar.

BTW, I'm still getting used to Spitball's grain prep tek.  Those grains look dry to me!  Following the Cult advice of following a tek to the T several times before asking questions about it.  At least the myc seems to be jumping off the agar wedge, so let's hope there is enough moisture inside the grains.  Time will tell.

Thanks, Tiger.  New processes always feel weird, but what felt "right" didn't work, eh, so this is the "new right" :laugh:  Looking forward to a healthy jar to take to bulk soon.

I use it with Pan Bisporus to help differentiate cultures after germination on lme.

For example, I took several transfers from MS plates and moved them onto lme to let them grow out a bit and get some nutrients.  Then to water agar.  I took the fastest four from that and transferred one more time to lme to get my mother plates.  From there to grain to test out each culture.

It also gives me some peace of mind but mostly it's because I have yet to see bisporus look like anything other than mold.  So, instead of watching morphology, I watch for how aggressive the culture is when it comes to searching for nutrients.
