Agreed. The walls make it hard to place the transfers and impossible to stack and fit many in a SAB. I find that after cutting a piece you can stab it then kind of wipe it on the new plate. It "usually" works. I'd like to use petri dishes but I'm somewhat of an environmentalist and I can't stomach the idea of creating all that waste. Personal problems, I know.

I agree with the single use petri issue. I wanted to go with glass petris but the cost of quality ones prevents me from going that route. There are PP petris that can be sterilized in a PCer at 15 psi. My only attempt using no-pour in them was a little shaky, but I know others are having success with them.

Im doing pour agar. One interesting thing is this Watkins food color does not survive the PC cycle very well. It usually goes into the cycle brilliantly colored, and comes out barely colored.

My progression of recipes:

recipe 1:
400mL Water
7.2g agar agar
6.4g LME
1.6g dextrose

recipe 2:
400mL Water
4g agar agar
6.4g LME
1.6g dextrose
(notes: too soft)

recipe 3:
400mL
6.25g agar agar
6.4g LME
1.6 dextrose

You could skip the dextrose and see if that clears up your agar. The ME by itself will grow everything you need it to grow. You might try skipping the food coloring also, if you want clear agar. That's probably the most likely culprit, more so than the dextrose.

"organic" food coloring doesn't survive the temps in the pc. Just use regular ol food coloring.

I felt the same way. In my first 8 months I threw out close to 500 plates. I since went to this tek and use the plates he recommends. They are I slight bit harder to see through but I think it's a good trade off. I have 60 of them and keep them in rotation of about 30-40 per PC cycle. I've also put about 85% of my cultures in long term water storage to reduce the plates needed. I recently was pointed in the direction of condiment cups that have pp5 lids with the cup that I just ordered. I don't know how they are yet. Stone-producer myc is always so funky.
I love the color change, it's especially visible from the bottom.

Is this a jack frost trait?
Never had a germination plate looking like this.
Even the T1 is thick and aerial overlayed...

Some AA+ T2 plates from fresh start with syringe. These are one week after transfer i dont know if thatΒ΄s slow or fast. They look fine to my untrained eye, which leaves me to believe my issues lie in grain prep or inoculation. I have yet to put this AA on grain, i hope i get better results compared to my unsuccessfull KSSS grows.

I use those brown lids, wide mouth version, no gasket supplied or required. I think they are amazing, you can drill into them and screw in a syringe filter if you like, they will also accept a self sealing syringe port. I drill one 1/4" hole and rtv sealant a 1 1/4" diameter sfd overtop. Lids seal up great and after 3 years of PCing I haven't lost a lid yet.

No, just agar and a piece of wood. It's an enigma type mutation off Melmak TP. Extremely aggressive fruiter. It's actually one of the pics in my sig
