*New Guy* If that's not obvious... any ways I have several I'm working with I'm trying to learn from. I'll love any constructive criticism given. I expect the worst.


this is "stargazer" I received as an Isolated Spore Syringe I grew out on agar then tried to isolate Rhizomorphic growth and ended up with this. i noticed the stargazer always seems so look almost "spotted"?



here is more attempts at isolating B+ rhizo. I have a plate of where I took it from. It seemed like the more I tried to isolate this strain the less Rhizo would pop up....Maybe it's watering out the genes????


This is me testing out an LC I made out of the stargazer ISS to test for contamination before innoc. millet jars.



This right here\/ I'm very proud of "mexi" i germed from spore print this is the second transfer and it's trying to reach out for the heavens but I see contamination and sectoring.. I didn't know if this is going to effect me trying to grow this plate out for later inoculation????


This last one I think is the ugliest thing ever. With that being said. I had a healthy ape jar I was spawning and thought "Oh I should grab some genes before I throw it in the sub. So I put the grain on the plate in the SAB but it was not center at all. So I played this stupid unintentional game of roll the grain on the plate for a while. I'm just trying to keep the genetics going on... it looks like mycelium is eating most everything else. Do you think it'll grow out or trash it?????

I'm using 1/2 pint jars with unmodded lids for agar. Do I do the same as with grains? In other words, should I flip the lid and leave slightly untightened for GE? Or can I put the lid on the "right" way and seal tight with no holes? Not sure how much GE (if any) agar plates need.

They need oxygen, but should have enough in the jar with the lids tightened down.

What kind of lids? Unless you use silicone gaskets, they wonโ€™t be air tight, even tightened down.

I guess it depends on what you call a seal. I mean that there will still be some gas exchange for O2 in the same way that plastic wrap around agar plates still allows gas exchange like Parafilm so the seal isnโ€™t good enough to stop mycelium on agar or prevent it from pinning, etc.

Saw someone mention reviving dry mushrooms. I wanted to try, so scraped some gill tissue from dried Averyโ€™s fruit onto a couple agar plates. Lotโ€™s of growth in just a few days. I donโ€™t know if itโ€™s just spores germinating, or if the dry tissue was resurrected.

Looks like germ plate imo. Prob a mix of both ms and cln growth.

ive cracked a capsule of ksss someone gave me open and put a bit on a plate.  made a real
mess but i eventually got a clean culture from it.  it was good experience.

I learned tonight that being in a rush and doing agar work is not a good idea.

I am headed away for a week and wanted to get grain started and do transfers of HBOโ€™s Ghost and GT plates that I have.  I am fucking everything up and there is no time for a redo.

O well.  Hopefully the plates dont get whacky in a week in the fridge.

The regular lids that come with ball mason jars with the rubber perimeter. I assure you they're air tight. Why else do you think we crack the lids when PCing? If you don't think so go seal one up and PC it. LMK how it goes.

I think she was assuming you were using one-piece plastic lids. Those are commonly used and highly recommended over the two-piece metal lids.

With that being said, I really dislike using jars for agar, but they can work. Mainly, I just didn't like the steep angle it forces me to use for transfers. That makes it difficult to get the agar segment placed in the middle instead of stuck on the tip of the scalpel or knife.

Yeah the plastic Ball brand lids have a plastic bevel in them that works how the lid on a pyrex media bottle does. I put water in a jar screwed a lid down tight shook shaked jiggled for a bunch of mins not a drop came out and after opening it was dry beyond the bevel. Guess I'd say good to go? :shrug:

I used three drops of food coloring i wish i would have only used 2 or 1. Less is better for sure.

Ive been running into a rash of penicillium (white with green center appearing later), but I think Ive finally gotten a clean culture of something beside cubes.

Im experimenting with using less agar in my recipe for a softer result as I was having trouble getting spores to germinate, and so far the myc growth looks better in general on these plates, even though they look kind of unclear. Potentially my LME is caramelizing?

Are you doing pour or no-pour? I doubt it's caramelizing, as I've ran no-pour MEA for 45 minutes at 15 psi and they are still clear. What recipe or percentages are you using?

I'm a fan of softer agar. I think it should be as soft as possible and still be transferrable, and not only for germ plates either.

That transfer is looking good though!
