Am I the only clumsy idiot who has ever knocked a stack of freshly poured plates into his flowhood? 🤪

If not … how did you clean the agar off your filter?  The damage in this case is only an area of about 2x3 inches…

I’m thinking I will get some near boiling hot water in a spray bottle and try to spritz it way. Maybe follow with a solid dousing of Iso. Maybe a good spray of Lysol to seal the deal. I definitely don’t want to scrape or scrub on the filter.

Thoughts?

Some agar problems are hard…

A clone is from this batch of B+.

They were thicker and had wavier caps than the other 3 tubs that round.

I’ve never gotten APE to germ on my streak plates. I always have to jamb the swab in the agar and it germinates around that area.

I figure I’ll give it a couple weeks. Then I’ll put a couple drops of sterile water on the swabs and streaks and give it another couple weeks. After that I’ll be begging for someone to send me more swabs.😂

Would have had a higher probability of success by pulling cotton filaments with your scalpel and burying those into multiple plates  but fingers crossed.

 I do recommend dipping the swab in sterile water first though. It makes the cotton much easier to separate and hydrates spores too.

I use 8oz jars to PC my water but no pour containers would work too.

Since it was just sitting in the lab fridge, I diluted some leftover agar. It’s in the PC now. I figure dipping the swabs in sterile nutrient juice before shredding on the plate is superior to just water alone. Thanks again.

I'm new to agar. I've done about 20 jars (1/2 pint) of all different kinds. BRF agar, LME agar, potato flakes/corn syrup agar, PDA. Inoculated via MS syringe, transfers, steaking with a loop, G2A, etc. Every single time the mycelium just seems absolutely intent on growing upwards rather than outwards. Am I just not worthy enough? Please educate me as to my blunder!

Another thing I find strange given my dodgy track record and being a noob, is that not a single plate has contammed on me. Not even when a drop of soapy water from the ceiling of my SAB fell into one of the plates while doing transfers (never misting the ceiling of my SAB again, only sides from now on). I've shined a high powered flashlight under the plate and it all just looks like solid white healthy mycelium every time.
