The fuzz I had transferred was apparantly trich. This happened overnight.

Still an interesting experiment. Going to try another tek to try to revive the dry sample

I’m on T2 of HBOs ghost. The original germ plate looked quite good I thought and I’m now on T2. These look to me to need at least one more transfer.

They came from these donor plates.

I am wondering if I should take another transfer from the top left plate at 2 o’clock?
Ghost is a first for me. The myc is growing very differently than all other cubes I’ve got. Are my plates just still fucked and needing more transfers? I still don’t really know what I am looking at other than when myc looks completely uniform.

you have plenty of good rhizo spots to take transfers from. I would do like 5 transfers or so from that plate starting at 2 o clock and going down .

Yeah that sector at 2 looks the most uniform n vigorous. I like clean uniform rhizo growth too before I send, though tomentose doesn't mean bad. Rhizo is just prettier.

I am right now PC’ing my agar media bottles without having loosened the lids. What a dumbass. I took them out of the micro, shook them and placed them straight into the PC and closed the lid. I just realized I never loosened the media lids again after the shake.
Is there anything I can do to save these bottles at this point?

Yeah, I would remove them from heat immediately and fix it.

Interestingly, when I pulled them out to loosen the lids, the agar inside the bottles was boiling - was the agar hot enough to be sterilized? Also, I use the corning media bottles and the lids are always a bit expanded due to the heat - when I tighten them down after a proper cycle, the threads are definitely expanded at least a bit.

I always PC media bottles (and everything else) with the lids on tight, no problems so far. I believe RR did too.
Correct me if I'm wrong, but the water inside the bottles boils and pressurizes them, so they don't implode. And if pressure inside the bottles increases the boiling point also increases, so the water stops boiling and the bottles don't explode either.
Provided you don't vent the PC of course.

That’s very interesting. However, it messes up sterilization tables that assume all air has been expelled during a venting cycle.

Amazoninan tissue clone plates here. This is t1, one transfer from the cloned tissue. First, would you all call that a t1? Second, I am trying to make sense of the two plates. One has rhizo growth that hopefully looks solid, it’s vigorous that’s for certain. Second plate (and there are a few like this) has half rhizo and what appears to be half tom growth. Is it common to see this variation from a tissue culture? Thank you.
