So 4 plates turned into 36 plates.  I got a feeling I will have more plates than time in the year to grow out.

It is a stainless steel straw.  I like that it has a bend in it, so I don't have to put my hands over the plate. 

I have 3, got them a while back for like $8-12 for a set of 6.  3 straight I use for mixed drinks and 3 curved I use for making transfer plugs lol. 

The plug that comes out is about 4mm or just over 1/8".

6mm is probably fine I have seen larger pieces taken.  That sounds about like the average of "eraser sized"  I think the process and sterility is more important than size.  Smaller just let's me take more precise samples, and more samples from the same amount of agar.

Not that I know what I am doing yet lol

It does take heat well.

I learned I will never get it red, so I just give it 20 seconds or so. The nice part is it holds enough heat it doesn't burn me.  I keep a sterile piece of foil (the one i used the PC the straw folded so the inside is now the mat) while I use the scalpel to move the transfer in my SAB to set it on ONLY after heating. It keeps me from needing to cool it in agar.

SymPlayTon: the straw idea is brilliant. Also, Iโ€™m curious how those 36 plates will compare to each other when grown out. Fascinating.

Sharing my current side-by-sides strictly forโ€ฆ pretty ๐Ÿคฉ. While there arenโ€™t enough samples to make any real observations, Iโ€™m using a potato flake agar recipe in one dish, and malt extract for the other. The nutritional differences arenโ€™t affecting growth speed anywayโ€ฆ

Next, when to do a second transferโ€ฆ a few more days perhaps?

They all look clean, and some of those look pretty uniform to me.  I would think you could use some of those directly when they finish growing out.  I am no expert though lol.

So make sure your wood fits on a plate it's a beech to trim it without it flying away.

I had been told here that it was there for more water retention, looks a lot harder to pull the wood out than scraping a bit of myc out of the slant.
But idk have never worked with them.

I pour 80 at a time. put tape over stacks of ten, put back in those bags, tape up, cover with clean shirt.

Let them sit 10 days room temp, makes sure none bad, stick in fridge the ones I don't use soon.

Technically it's possible to have contam that shows up after 10 days but it's rare. 10 days weeds out the one or two per every other time I do this that will have a contam.

Yup! Southern Labware. I had my eye on a different one, but I was having trouble getting them shipped to my location. Which was the concern of the user who suggested them lol.

Anyway, thanks for the workflow. Thatโ€™s what Iโ€™ll do ๐Ÿ‘

I have a lot of plates that need transfers soon. Iโ€™m excited.
