I have a question about this video. When the agar is stuck to the scalpel, he uses the lid to knock it off. Is this good sterile procedure?

I like the timpelapse of the one on red agar. You can see it growing weirdly due to the red dye #3

I wouldn't use the lid cause it'd add a possible contam vector. Better off practicing your technique to get transfers off the blade by practicing how you get them onto it.

I saw the same thing.
No that is not a good practice.

Interesting video none the less.. I was definitely surprised with how plate 1 turned out.. it has me thinking about adding more diversity to some of my transfers to see what happens... I don't think I'd have ever taken a transfer from that first spot...

I also liked how he numbered his transfers so he could go back and see where each one came from... I've seen someone do this before, but I didn't really see the practical application for it...

Thanks for sharing the video shroomsmurf.. I quite enjoyed it..

Too bad he didn't put that last one on one of the blue plates... I bet it would have done pretty well.

Erythrosine is the red dye #3 so any color that's made using that color wouldn't be a good choice either. RGB is the base color palette. Anything from the GB spectrum is fine.

When using agar to "clean up" multi-spore sources, how long do you let each transfer plate grow and what is reasonable regarding how many transfers to make before going to grain?  Asking because I've seen people suggest immediately going to another plate when the myc is no larger than say, a quarter, and I've also seen folks wait until the myc is almost taking the entire plate before doing another transfer.  Also, if T1 looks clean, is there any reason to take it to T2 or further?

Best to transfer from a germ plate as soon as there's growth so as to get it before anything else overtakes it. Then you let that T1 grow out more if it's looking clean. Even if the T1 plate looks perfect your going to goto many more plates from that one to expand the culture family tree wider. Plus going to T3 or T4 plates before grain gets you a little further away from pure multispore genetics.

Thanks for the quick response.

In the specific case of Ps. Natalensis, where the genetics, in my opinion are much closer to wild, than most actives that have been cultured for so long, and there appear to be so many phenotypes,do you recommend the same?

I don't have experience with Nats to answer that confidently. I imagine yes, I'd do the same. But I'm not sure. It seems like Just a good standard technique approach for all varieties really.

I found these on the big A site. Would they be good for agar work? Lid and cup are both pp5 so I'm assuming they are. What's the best tek for these? I was thinking pasty's but was also thinking about putting a filter on the side so I don't need to cover each one with foil.
Here's a link if you want to check them out on big A site. Hopefully I'm not violating any rules by posting a link, if so let me know I'll remove it.
