It depends. Either way can/will work. However, "fast-transfers" are often employed when trying to outrun contamination(s). I've not been following this conversation, so what is/was the source of your spores?

Really just depends. You are pursuing quality. My very first run took to T5, and even after that there were still iffy plates. However, things got much better by/after the T5s. Don't bother getting in a rush. The tortoise generally wins this race...and it's worth it, IMO.

IMO, since it's a T1 I would suggest some fast-transfers - generally about every 4 days. Transfer as little myc and agar as you can to help clean it up. I keep the older ones to see how they grow out...and for the plins (plate pins).



Right that terminology makes more sense, thanks.

I bought a spore print from a vendor and scraped them onto my germination plate.

When you say that one will fast-transfer to escape contam, does invisible contam exist? As on this plate I can only see one bacterial spot at 3 o'clock.

What I mean is I have other transfers with no visible contams, should I be transferring those for the purpose of genetics selection/monoculturing? Or am I trying to get rid of invisible contams?

Sorry if this is confusing, thanks for the help.

Ok gotcha. So I'll transfer in the next few days from the one with a spot of bacteria, but for the seemingly clean ones, I'll wait a while longer to find the best mycelium.

Dumb question...if I pc some plates but want to wait to use them. How can I keep them sterile for when I need to use them?

I finally have a little growth on a plate and want to give it a little more time to take the first transfer.

Washed some WBS last night, is soaking now.
Here's some T1 xNipsX headed to grain as F3. Also some clone T2 of the non-Nip big thick x7x mutant that's in my avatar pic (16 quart tub). This plate was was in the fridge a while, then thawed and started growing a little.

How do these cultures look? They were all inoculated from MSS on 7/27, so 8 days old now. MEA+Yeast. The one at the very bottom looks like contamination to me but that's the only bad spot I see so far out of 25 plates so not bad for the first time.

Yeah bottom one looks wrong. Lotta good spots in the others to take some transfers from to put on new plates.

The bottom right one looks kinda iffy also

Hey can someone give me some help here? I had a small amount of rhizo growth on this original swab plate that I made some super tiny xfers from. I was low on plates so I put 2 -3 xfers on the plates. However, I feel like something is off with these. They are SO fluffy looking on regular 10/10/500 mea agar. I know I could lower nutes, but do you guys think these have some ride along bacteria or mold?

And here are the xfers grown out a bit, at first they looked a bit better I think but as they've grown out they've gotten very fluffy looking. I ordered some gentamycin to try to outrun bacteria if that's what is it. I also did a Josex poke to BRF puck.



What do you guys think? Are these clean and just super tomentose looking or do they have something riding along? These are my only cultures rn so I'm stressing it and don't want to waste my time if I should just be starting over.

They were T1s that were transferred from a multispore germ plate on 5/18, about 10 weeks or so.

The germ plate definitely isn't perfect, as to be expected. These ones look better, they look tomentos for sure, they may still be carrying something, but you can definitely keep cleaning them up. I'd let them grow a couple more days depending on speed, then take more transfers again from the cleanest looking organized leading edges.

Ugh that's what I suspected. I don't understand why I'm having such trouble getting a germ plate that looks good enough to produce plates without these "hidden" contams. I never see satellite colonies, it's always mycelium like this though, all fluffy and looking like it's harboring hidden contams. I don't get how people have these t1s that look so clean. My initial plates always have something intertwined

A lot of times people make lots of germ plates at once. Putting spores to a whole sleeve of dishes at time. It doesn't matter what your germ plate does really. Just keep transferring multiple transfers to multiple plates till you get something that looks uniform and organized.

Fluffy tomentos mycelium doesn't mean it has hidden contamination. Plates don't have to be all rhizo. Both types will fruit just as good. Some varieties dont produce rhizo growth. Just keep transferring and enjoy the process. It will work out. Your dishes don't look bad at all. I've seen people clean up waaaay worse lol
