Re: AGAR ENVY! (Anything and All things agar!) [Re: Tormato]

Hey so have an lc working and tested on agar with sterile swab I don't see any contam spots not sure of how the pics came out but and thoughts are sporeciated

Also I know I should already know this but I've been wanting to be more discerning when picking transfer spots from a plate.  Does anyone have links or video to help?  Summertime high temps not consistent results so starting to analyse each step of my process and find where I can improve or step my knowledge I guess sorry not very good at expressing in words.  Thanks lol thanks

I pull out the alcohol wipes, Clorox spray and Lysol.

mixing alcohol and clorox can produce toxic gas.

Lysol is sticky, smells horrible, and leaves a bad smell for days.

Skip clorox and lysol and just use 70% isopropyl alcohol in a plastic spray misting bottle. That's what almost all of the people doing good work here use.

70% works best, not 90%.

Just wondering if the white growth even looks like mycelium?

There's some penicillin mold growing here. The big powdery white patch turning green on the left of the left one, and the little greenish circle on the top right of the right one.

Looks like you transferred from the right place, if there's any clean to transfer from. Not sure you'll get something clean.

I'd throw these out without opening again. Once penicillin is green it's spreading spores. You do not want that in quantity like this in your home.

Work your transfer, but also start over after reading up more on basic technique. Good link to good teks in my sig line.

There's also red bacteria colonies on the far right of the right one.

Your agar looks good though. Which recipe did you use?

Thanks for the reply, cool that it’s penicillin . The pink stuff is also very interesting looking.

My transfer is growing nice white stuff and the original plate continues to look more confidently like mycelium. No contams after 36hr but we will see.

The plates are pre poured. I would like to make my own if there is a good tek without a PC, but for now I’ll minimise my variables.

I appreciate the help and will work on some new spores

I made my first batch of agar the other day and think it turned out pretty well, I inoculated using Tidal Wave MSS, Mazatapec MSS, and a Pink Oyster spore print that I took myself off of a kit I bought a while ago.

I do have a question, some of my plates have these dark spots under some of the air bubbles as seen in the TW picture, they're definitely not on top of it. (there's a little myc growth there in the middle too ) What could those dark spots be? It doesn't really look like contamination to me but I have no idea.
Agar recipe is ME+yeast.

Am I correct in assuming that if a plate goes from really ropey to growth that looks tomentose like this one, it's probably due to contamination?

If so, any idea what type of contam it is? Is it possible to clean it up or is it the lost cause I think it is?

Any chance the donor and receiving plates are different nuts?

Infected or not is inconsequential, I think this is just kinda neat.

This is a T2 from a Redboy Revert print that has been going all Billy Idol.
I came in the room and it screamed more more more.

Yes... it's enigma, so the donor plate was sent to me from another cultivator. I don't remember exactly what notes are in his plates, but I know it's different than mine

Loving the labels, sexy. I have a DYMO 4XL, I deal with so many plates at one time that it would definitely be a lot more hassle for me to write out and print labels. I was labelling tubs at one point but the novelty wore off pretty quick.

Looks to me like the mycelium changed growing style when it reached the new dish nuts.

Awesome.. so it still looks clean, just switched to tomentose growth?

I'm going to try doing another transfer to YMEA tomorrow to see if it does anything different. I'm pretty sure that's what the original plates were.

Obviously I cant say for sure but I think its fine. Id do exactly what you describe and try a different recipe. My current APE culture hates LME for some reason.

Haha  yeah I love the little printer I got it’s called NIMBOT
i like that it has an app and I premade those labels just change some parameters and print. They also come off pretty easy and this way I keep track which plate went to which jar and ultimately the tub

penicillin is a contam if you're growing shrooms, and it's common.

I used pre-poured for the first few runs I did that worked.

Your pre-pour plates aren't peptone are they? That's commonly offered but increases chance of contams. Is for growing bacteria in labs.

When you get clean plates with pre-poured, this agar works well for me. Also some tips there that may help anyway. You can skip the charcoal at first.
