I think a stick blender goes a long ways
to getting rid of sediment

So if i mix it well, with mixer or blender next ones should be good?
Maybe it’s also cuz i store lme and agar in the fridge and without letting the temp down and mixing with hotwater ?

I've only had sediment using pasty's plates perhaps from pouring too thick. I switched over to plastic plates, pour as thin as possible to cover surface, haven't had problems since.
btw I'm using LME, I mix the lme with agar powder add water, mix well with a spoon, and then just get to a boil (imo helps even out consistency)then pour into container and PC

The stick blender you can use as it simmer
cooks on the stove. I put water in a quart
jar, then quart jar in a pot of water, heat
it up slowly, add in LME, low simmer and
stick blend. The quart jar helps with boil
over chance and allows the stick blender
to be submerged nicely. Then i add agar,
and coloring. Then stick blend a bunch more.

Toss a grain jar lid setup on the jar
and PC 15psi 20mins, or funnel into
a media bottle, or do a no pour.

so I had some interesting transfers from this manky dish:


these two are the best, taken from the 9 o'clock section, but look like they're enmeshed with something? random mold in there as well do you think? I think the plates are pretty clean.



really appreciate all your feedback folks, it stops the ole head getting too anxious about simple stuff, ya know?  :laugh:

All I see is mycelium, others may disagree. 1st pic I wouldnt take to grain though, could use a transfer or two.

I just used 2 syringes and a print to finally get back into agar work after like 5 years. Both syringes and the print are pretty old so let's see if I get any growth 🤞🏽

I would recommend it unless you have a very steady hand. I wrapped my first set of dishes in open air, most of them got contamination along the edges when the lids wiggled

I wrap in front of the flow hood, but how do spores in the air work their way against gravity and get between the lid and the dish and to the agar?

When wrapping plastic plates, it is easy to squeeze them in the middle, which sucks in air around the edges. I unwrap and rewrap plates in the SAB, always.

Contams can be sucked in on this airflow around the edges.

Another question... Once it's on does that make it ok to pick them up in open air to check them out?

Yes, once wrapped, they are safe to take out of Still air or flow and store

Yes again, the parafilm protects the inside of the plate from contamination.
