That's great! Will you post the results even if you don't see any difference? That's an important result in itself.

Sounds cool. I'd be interesting to get a comparison of different mediums and their effects.

I had some germ plates that I put a swab to about 6 weeks ago.  Nothing every came up on them - can I use them for transfers or maybe to cool the scalpel? They look totally clean but I feel wary for some reason.

I had a dream there were clouds in my coffee.

Transferred a bunch. only a few shown here.

Day 9 (date on one plate is wrong.)
75 degrees natural heat.

I'm running a nutrient variable experiment right now.  Spent time over the weekend painstakingly making individual no-pours and precisely increased the amount of nutrients in each one.  The baseline was 10cc of water in each no-pour and 0.2g of agar.  Varied the LME from 0.5%, 1.0%, 1.5%, 2.0%, 2.5%, 3.0%, 3.5%, 4.0% and also BRF at 1.5% & 2.0%.  Tonight I carefully moved over the 10 transfers.  The dishes were slightly soupier than I would have preferred but I'm still really excited to see what they look like next week.

Will post a thread if it ends up panning out.

You know, right, that to produce any useful data from a "change one thing only" grow require using single culture clone selected down to a single sector and grown out.

Without that, there is too much variation possible with a multispore grow to really determine anything.

Yeah I don't actually plan on publishing this one to the Journal of Mycological Sciences, maybe next time though.  In my home lab there are obviously many other variables, sector selection, wedge size, accuracy of my measuring devices.  Regardless it will still give me a sense for what works better with the products that I use.

I don't know who decided that nutritional yeast wasn't a necessary additive to MEA, but as soon as I included it, my growth got way healthier and faster. Up until then, all of my MEA plates were thin and wispy. Now I'm getting strong rhizomorphic growth.

Nice multi! My first cloned specimen are finally showing growth. It’s wavy capped Mazatapec. If it goes good, I’m going to store them and look for something leucistic to cross with.

Working on a couple Pan projects.
Wild South Carolina cubensis
KSSS - this dish looks like a head dress
RW - ready for transferring to grain - Notice in the 11 oclock position some slight Rhizo activity. People say that RW doesn't produce rhizo's.
PE6 - ready to send to grain

I apologize if this is the wrong thread but is definitely agar related. What is this pattern of growth due to?  All of my T4 dishes look like this from MSS to agar.  Symmetrical growth and no obvious contamination that I can see but the immediate 1 cm or so from the transfer wedge looks different than the rest of the growth. Is it something that I should transfer an additional round before going to grains?  Thanks for any advice!

Idk why that happens sometimes. That plate looks clean to me.

Just a theory, but I suspect that it happens to me when my agar has some condensation that has not reabsorbed yet. The myc grows like that when it is wetter and then forms more nicely once the moisture is more ideal.

Could be pure bs, though. I have a few fresh plates right now that I can detect a bit of surface moisture rolling around on if I tilt them too much. Otherwise it is pretty invisible. But in a few days, they will be dry if they follow the same pattern. I will try to grab pics when I get home.

On the 30th I did my second run of agar using the clock streaking method. So far, there's nothing visible on any of the other plates except these two.

Amazonian Print

I'm totally baffled by how this even happened. I have no idea if it's any good or why it even grew this way. The little sliver line on the edge above the writing is 12 o'clock. Also, this grew rapidly all things considered.

A.P.E. Swab

Then there's this. There's obvious contamination on the side there. I think I see something near the swab itself, but at this point I'm conceding that I really have no clue what I'm looking for in the first place. I'm not sure if I should let this sit awhile longer, or if I should try and grab a sample from near the swab where a little tendril has grown out.

I still have plenty of clean untouched plates from pouring. Of the stack, it appears one or two are contaminated just sitting in my SAB with the arm holes covered. I feel frustrated since I haven't even touched anything in there since pouring. I'm not sure what I'm doing wrong.  :uhoh:

Any help would be appreciated. Thanks for looking and reading.
