Ok yeah but you have more healthy looking myc trying to grow. Those Yeti plates are 2 transfers from the Swab Plate, and they look like that with that huge halo. Doesn't the myc look unhealthy too? I mean I could just keep transferring it, I thought those big halos indicated something off. How come D3monic and all these guys plates never have that ?

So keep transferring the yeti you guys think? I'm kinda expecting it to do that "scalloped" growth where it's like super sectory and contamed, that's what's happened to a number of my plates.

I know how to transfer away from bacteria or some outside colony, but this myc just looks so weak compared to other peoples plates I see on here, even t1/t2 plates look so much better than mine.

Also on some of the GT plates that I didn't post pics of, literally nothing is growing except the "halo" looking part. No white myc at all except on the agar transfer piece, the plate is growing straight "halo", which I'm guessing is some contam since it's stopping the myc from growing. I'm gonna keep xferring the Y plates, and see what happens. Maybe it's just the growth I can't see, but some of the plates are certainly contamed from that "halo" looking stuff. It kind of turns a more solid clear and the myc barely grows at all. Thats the GT and the Stro (same source for both prints) though, not Yeti which are pictured here. Hopefully the Yeti are going to work. I wish I could get a print that was somewhat clean, I've tried many times and gotten many prints/syringes that just provided absolutely nothing transferable.

I can only speak from my personal experience but heres an example APE with "halo"
1st pic is ms plate, 2nd pic is first transfer and last pic is my t7 plate i put to grain.


D3monic is an agar wizard you cant compare ms plates with cloned cultures, dont give up yet :heart::mushroom2:

Try tweaking your agar recipe. 


I’m not a big fan of hard plates but it does seem to stop a lot of weird stuff from happening. I’d try adding a bit more agar, then if it keeps up try changing the nutrient.

Ok this gives me a lot of hope. That looks similar to my Yeti plate at least. Would you recommend taking a xfer of the 1st and 3rd pic now? Or should I wait a day or two? I feel like the 1st plate looks better than the one in pic 3, should I even bother with both plates or take a few xfers from the 1st one?

I always do quick transfers when I'm trying to get stuff clean.  That leading edge, near the halo, I would transfer from that with a pretty small piece. Like a grain of rice.

I would transfer from both of those plates and I would do multiple transfers.  Agar is cheap.  Plates are cheap.  Losing a month of work because I didn't take enough transfers feels bad man.

Also, you can put multiple transfers on one plate.  Take three transfers off plate one, for instance, put it all on one plate.  Then do it again with another plate, and you improve your odds of getting clean growth without a lot of work.  Six transfers, two plates.  Do that with both plates, you got twelve transfers and only four plates to worry about.

Should give you something clean for sure unless your sloppy.  Which happens to all of us btw.  I still get in a rush and all my clean shit turns up bacterial if I'm not paying attention.

IMHO
If it were me id transfer from both plates. Tomatose growth isnt necessary bad and I feel like having few extra plates will only increase your odds plus good practice.

Ive taken transfers from one plate days apart from each other because I saw new growth that looked good. So its up to you. You could take one now and maybe in a few days if you see some better sectors on the original plate take another.

With that being said as long as your agars clean there is no harm waiting a little longer to transfer.

How many transfers do you guys do from cloned tissue? I would assume at least one, so that you aren't innoculating with mushroom tissue mixed in.

I've been putting tissue to agar, then transferring from the healthiest, fastest growing sections if the growth is uneven.

I've also used a couple clone plates that looked super clean and havent had any issues as of YET, but, it's probably much better practice to just do at least 1 transfer

Yeah, in addition to clean samples, I'm looking for fast, healthy growth as well. Sometimes a clone culture shows uneven sectoring on the T0 and even T1.

Most of my clones sector a ton until like t4.


I rarely make it that far though, t2 is usually enough isolation for me.

I don't see a reason to keep transferring if your growth is clean and uniform
I just used a t0 clone plate to biopsy into an LC the other day :shrug:
maybe I'll pay for being lazy but only time will tell

I think it's the same idea as transferring from the leading edge from MS.

Granted if you get a clean biopsy, I guess it doesn't matter as much.

Lately I've been fucking around with cloning tiny ass tamp pins and I plan to make several transfers.  The last clone I took was from a sclerotia and I only transferred once but I could have gone straight to grain had I wanted.
