What do you guys think of this. I can’t tell if it’s bacteria or not because the only bright white myc I see is coming out the top of the spore clumps

Nice notes! I might have to copy some of your spreadsheet style.

I'm mostly keeping track to figure out where I've gone wrong (In the past) and trying to tighten up the whole process. But does it not make sense to keep your old culture plates that haven't been stretched too far, in case it was a bomb culture and you want to store it long term, or want to start another batch without using spent/recessive genetics? Also to compare my future work to in case I've forgotten what I'm really looking for.

I've been on a losing streak lately and I figure more detailed notes will clear it up.

well i guess this place is as good as any to post this of some transfers from jakes nec-d pan these were all taken only a day n a hlf ago off a barely germinating streak plate. i just cant get over how fast these things are moving. plus they look good

Man I just got a print but now I’m waiting on my case of plates before I can get it started.


I’ve had a couple take off like that but it’s crazy they all did.

To me too.

Also, 2% agar and 2% LME is the gold standard, that means 10 grams agar and 10 grams LME to 500ml of water. It's fine to go lower than that but 2% nutes is not "a lot" for agar and I personally prefer it over lower nute strengths.

Maybe you'll get thicker growth if you add 8 grams of malt to those 400ml of water.

I’ll make a couple bottles to try next time I make agar lol. I’ve had bright white growth with this recipe before, but this is two transfers out from an iffy pinned plate I made in June. That’s why I was wondering what people with more experienced eyes thought

Where would you transfer from on this plate?

I could grab an individual sector but wouldn't that further isolate the genetics to some extent?

There was a thread on this a few weeks back and the general consensus was that it's not going to drastically affect the genetic outcome unless many transfers were made in an attempt to further isolate.

Any thoughts Agar gurus?

These were done with slightly less (1.5% agar/LME) as an experiment but the next ones i'll do at 2%. Is that considered to be a general sweet spot for agar / LME?

I know it is early but Im wondering if I should be looking for any characteristics that would let me know if any of these could be good candidates to go to grain or if they need more transfers. Want to make sure I'm not rushing anything or being overly impatient but also don't know exactly what would qualify something to be ready.

I can't tell if the growth on the first two (or any really) are going well enough to go to the next step but these are what I think are the best ones!

edit :: looking at this I realized I took the original sample in the transfer. how could that potentially impact things?

I bought a cheap light box. Pics are looking way better:



There is still a bit of glare on the edges of the plates and the sides of the jars, but it isn't awful.

Yeah cuz with pans the type of myc is important so with all those wedges there’ll be plenty to pick from. In this case the pretty much all look good but that doesn’t usually happen. They won’t get grown out much I imagine he’s probably taking the next wedges today if it’s not already done.


Lol it does look crazy with so many though.
