Noobie here,

So on the 24th of August I inoculated some plates using a 1.5 LME agr solution. i made up 7 plates. Knowing I forgot to PC them i dumped four of the seven and re-poured agar and inoculated again. On the plates that were not Pc'd I have this almost wispy white growth. The ones that were PC'd have a bunch of contamination. By logic that doesn't make any sense to me. And the white growth in the un-sterilized jars is not showing rhizomorphic growth as far as i can see. I have two of those plates that clearly show the growth on the path of inoculation. So, I'm just looking for some advice on what to do.

So, wait, your "sterilized" plates have nothing but contamination but the unsterilizied ones have growth?

There is no question that the sterilized plates are contaminated. I'm not going to take pictures but, am going to wait for another day or so for rhizomorphic growth. I guess I'm just looking for more tips in sterilizing using a pressure cooker.

I mean the other plates. If they weren't sterilized then they are probably also contaminated.

Make sure you are venting your PC.

And what do you mean by venting? I have a pretty standard pressure cooker that just keeps it at 15 PSI.

Bring it to a boil without the weight on. When you hear it start to hiss leave it on high for atleast 10min. Then put the weight on, let it start to rock and turn it down to just above medium so it maintains pressure

That helped my bacterial spawn problems a lot. Any bacteria i get now i chalk up to poor technique

Awesome, I will definitely try that technique out. Also, about how much water should I use?

Weird deal but the temperature is not high enough without doing that.
Something about steam vs air.
"If there is air still in the vessel then the temperature will actually be lower than the pressure would indicate. It has to do with a scientific principle called “partial pressure”.

Great then I’ll transfer one more to save genetics. Would you recommend dropping the hole dish in 1 jar or is it better to diversify?

Also wondering what the process is as far as picking agar mixes such as PDA, MEA, MYA, PDY according to where you are in transfers? For example do you prefer to drop spores on PDA then changing to something different or do you go T1- T?? and change the mix? Was recommend to drop spores on PDA then move to MEA after T3 but can’t bring any truth it...

A few grain sized pieces of the leading arm between 9 and 11 might do well. Malt extract is recommended for all transfers. Potato is often slower than malt.
