I don’t use foil anymore, just leave the cap slightly loose and pc. I’ve never used one of those filtered lids though

One thing to note is that ptfe filters usually wear out eventually, for grain jar filters I think it’s around 30 runs but that could be different for you since agar doesn’t have to be sterilized as long

Yeah, I might end up not using these filtered caps. Never had problems at all with the low-tech methods. Just was curious.

God point about the wearing out of the filter ! I have not thought of that.

It seems to be so. I just dropped, aa, peu, ape, and some Columbian rust spore on agar. I am trying a new agar recipe basically cuz I ran out of lme. 500ml h2o, 9g agar/10g potato flakes and I was going  to put a few drops of honey in there but totally forgot to add it to the mix before I PCed. So be it I'll see if I can get spores to germinate on it, I've used potato soak water before with success, can't be that much different.

Exact recipe I used experimentally before switching to brf agar. I tried it with both honey and yeast and while honey added gave better looking plates,  potato flakes as my primary didn't perform very well (not as well as potato boil water for whatever reason, but all flakes I imagine are not created equal).  Wirked well for germination but gave weak growth.  Played with the amount but wasn't terribly satisfactory.

I agree with potato flakes working, just not the best..

That recipe sounds really rich.

I use like 7g per 500ml

Yea it was literally all I had available. My lme ran out and I forgot to order more, but I do have brf so maybe I'll find a use for it in agar.

Brf makes pretty good agar, especially the store bought, industrially milled stuff

What's your mix, I typically go 700ml h2o in my 1l flasks. I think I have some fine ground brf.

I use lme so idk. I try to go around 1.7% nutes and 1.7% agar

7g of the brf I grind myself or 4g of the store bought to 9g agar to 500ml H2O.  I mostly just use brf I grind myself honestly although I have a bag of Bob's  sitting around 3/4 full.  I  have a little strainer I use to sift the finer stuff out of my home ground but still need to use the higher amount even if I let it sit a while before adding agar and turning the heat on.  Both benefit from a little bloom time in the water. 
Afterthought/in addition: I don't decant or filter mine.  It has a few flecks of undissolved brf in it.  Decanting/filtering just isn't necessary.

These rings are an indication of?



What causes this type of growth on a plate? Foggy to rhizo.



A piece of grain got squeezed and bacteria seemed to ooze out, but after a few weeks it seems the myc is winning.

Wanted to show you guys/gals a nifty storage drawer that I’ve been using for room temp agar cultures.  You could fit 216 60mm petris, or 96 90mm petris, and allows you to easily categorize and sort through them.

Also did a big block of SAB work last night - 30 transfers Costa Rican —>t4 / 6 transfers Malabar —>t2 / 4 transfers Costa Rican —>t5

Took some pics of the prettier ones:

Temperature fluctuations case those "rings".  I get extreme ones due to 20 degree temperature swings.
