Was the streak plate from a swab? Was that the first work of the session?

streak plate was a vendor LC, I dropped a single drop from syringe to my loop. It was actually one of the last things I did in that session.

I streaked a blue oyster LC and Black Poplar LC

the BP didnt pink out like that. Loop was heated red hot, and I think I cooled it in plate prior to squirting a drip

Myc doesn’t like too much sodium. Some people that have well water have to use bottled due to water softeners. Trace minerals are okay I think but too much salt will kill myc

Good evening, my Agar Agar arrived and i have bought Malt Extract and 20 pieces of sterilise plates. Is there somethink i forget to buy? Is a sterilise room really necessary?

No, i do agar in my still air box 6 feet away from a moldy window

Do you have a scalpel/xacto knife?

You also need a pouring vessel. I just use a grain jar lid on a quart jar

1. Why do cultures start sending out fibers when they reach the edges of the plate, like in the photos below?

2. Would you transfer the below plates to grain? They are only the second transfer after MS (MS -> agar -> agar), but they look fairly circular to my novice eyes.

The labeled plate looks good, but the other colony isnt uniform enough to be just myc

They’re both labeled so I’m not sure what you mean. I marked them “1” or “2” in the lower left corner of the photos - which one looks moldy to you?

I thought it was split left to right, not top to bottom.

Its the top lit ones that look really disorganized.

Mught be bacteria, either way something is meshed in there. I would try a warm pour

The top lit ones are the same as the bottom lit ones in a given row, though (1 = 1 and 2 = 2). So do they look okay from the bottom? Okay enough to quell any worries when looking at them from the top?

Just poured my first stack of plates in front of the new hood and it was awesome. Im leaving them to get rid of the condensation on the lids, but I'm super excited about this new tool. Life changing experience, later im going to drop some ape, peu, and rust spore tonight its going to be heavenly to get out of a sab.

Yea the agar was hot I had a limited amount of time to get it poured but if I leave them in the hood it should be gone by the time I get back.

Hello Shroomery,

long time lurker here. Finally I have a question that I could not solve from reading older threads.

I used to do agar in all sorts of bottles that fit the PC. Opening stuffed with polyfill covered with aluminium foil.

Now I thought I would get a decent lab bottle. I got in addition to the normal GL45 caps this one here which has a 0.2mic filter built in:



Am I correct that with these I screw them on tight and cover with aluminum foil because the filter takes care of the gas exchange in the PC ?
