What kind of agar? I get little to no sediment in my plates with lme. Also once agar is PC I don’t swirl it or anything and leave the last 1/4 in the media bottle. That way any sediment that may be present stays in the bottom and not on a plate

grain water, oats specifically. plus, thinking about ordering the charcoal powder that you recommended because apparently the activated carbon that i bought is different? i have always thought that they are both the same thing... the one i bought is used for aquariums so im not sure why it doesnt work. i ran a test and when im mixing it, the water turns black but once i stop, the sediments settle at the bottom and the water turns clear/grey tint. it doesnt seem to dissolve and turn the water black

I like sediment. It gives me some contrast when looking at things up close under the scope. I've never seen it hinder or impede anything. That's just me though.

the more stuck!

I have a wedge stuck to the top of a Petri dish and the myc somehow reached across to the agar and started growing.

How do you do this, I tried and I ended up with one raggedy ass end piece just frayed to shit. Nowhere near neat enough for my liking 😂
My knives are sharp too.

Just save up and get parafilm. I fucking love parafilm

Wow! I just saw 76m of parafilm for £168 that more than £2 a meter.
Why is parafilm preferred over other grafting tapes? Or M3

Labgrade, allows the proper oxygenation of a sample, also has no odor and it's self sealing thermoplastic.

Basically it's lab approved, for lab work probably one of the few things you can use. Think I remember like a nescfilm or something that is interchangeable.

I just prefer parafilm over saran wrap and such.

Wish my SAT would look like that. Very weak looking. Need new print.

It was a numbers game. I made 7 streak plates and took 20+ transfers, this one was the beat but I want to say they about 60% of them looked average to questionable.  This and a few others look great though.

Your agar plates are living art. I know that sounds cliche as fuck, but you moreso than others seem to kill it with the colors and rhizo spread.

Post only the good and never talk about the bad.

I just threw out 4 plates that looked like I streaked the inside lip of a toilet. Can't for the life of me figure out what went wrong. Usually I know where/when I'm going to contam.

Hello all, here to learn some Agar techniques and trying to solidify some mycology in my fat head. Did BRF cakes about 10 years ago and then again about 5 years ago with mediocre success. This looks a lot more fun and proficient than shooting for success with cakes, so here we go.

In mid June ordered all the Agar supplies, gathered up all my old syringes (and a new one) and emptied them out onto 20 plates. Was around 50/50 contaminated to not (at least visibly.) It was probably late to do the first transfer, but I was out of town for a while. I poured 60 new plates yesterday and transferred to the new plates (not all of them, 2/3 samples per original plates.) Thanks to all for the knowledge here, I'm on page 40 right now and plan on reading all the way up to here I find a lot of great info on every page almost.

Here are photos of my OG plates. What is up with my food coloring, that was 5 drops of red into 500ml of water whisked in pretty well, all sorts of weird streaking, in a couple plates the color dissipated all together. Also I got a bunch of bubbles on the first pour but not later ones. Also photos of the bad plates; should I try to transfer off of those at all?? The one with the black spot looks to have healthy growth, the others look mildly interesting to me.

Wow brosiph. Those look good for plates inoculated with a syringe. Since you have so many decent germination points to choose from I don’t see any reason to mess with plates full of black, possibly deadly mold lol.

Myc eats the food coloring as it grows. Other stuff probably eats it as well. I’d take some transfers from the outer edge of a couple clean spots. I’d probably transfer into 4-5 new plates. Let those grow out. They should be clean but I’d still do one more round of transfers, but also drop a few wedges into grain just to get some jars started.

If you transfer into like 20 plates now you’ll just end up with a ton of overgrown plates of plates that get transferred from and never used.

Thanks for the advice! I really don't have a plan, I was figuring the more plates I do the more likely chance I will end up with something nice to put onto grain. Did you see any plates that are especially nicer looking? I won't touch the moldy ones, I have enough to throw them out. (Do you guys toss them straight into the trash?)

So now that I have 8 plates of t0. I made 2 transfers of each of those, so now have 8 t0 and 16 t1 plates. I won't make as many plates on my t2 transfers, but im curious what you guys do with all these older plates. How do you decide when you want to stop making new transfers?

Im no expert either but I usually start with oone or two dishes for spores to agar, then transfer out to 5 and maybe transfer those out to 10 or just use the. 5. Saves you a ton of wasted plates
