Looks good, did you clean those cups somehow or use them straight out of the package??


Yeah rice sized is perfect. I usually make a tiny triangle, as small as I can pick up. It’s probably easier to start a little bigger til you get used to handling the wedges. The smaller you go the less likely you are to pick up contamination and bring it with you to the new plate, plus it narrows down the culture more so it’ll be uniform growth much quicker ime.

So I'm up to T4 and average every 5 days to transfer.
That seems really fast, although I've heard Oyster grows fast.

There're still not very clean looking. And too bad the shark fin wedge isn't visible in TD4.2. LOL
Shark fin wedge = a wedge that lands on edge and looks like a fin.

Old plate pic kinda blurry but looks like healthy myc growing over the black mold . I remember that the mold was there and the myc grew to it


So would it be worth it to try and transfer sum of that . In hopes of getting a culture that fights off contams better like the natalensis strain...

10g agar, 10g DME, 450 ml water. Is this okay after 4 days? Seems a little thin. What are those white dots on mycelium?

Also, how do you deal with deposits, do you blend agar and DME? I just mix it with spoon.

D3monic mentions in his agar tek to use an immersion blender... lucky enough my wife is a pastry chef... lol... my agar turns out much clearer with a lot less sediment than ever before.

I've had various spores do that (white dots) on some of my plates but it does look kinda thin, but also seemingly normal for 4 days total in terms of growth rate.  What temperature are you keeping them at?

Makes a huge difference, that and getting the water to light boil before adding dry ingredients. Here’s some plates I poured last night

Depends a lot on your ingredients source too, obviously choppy fibrous agar agar going to give more fibrous results  same with nutrients. I been stick blending a good minute or so

Are your plates kept where there is no variation in temperature?
I'm thinking condensation is unavoidable with room temp swings.
Oh, and team telephone agar! :lol:

Does anyone use 'plant agar'? Agar sold for plant tissue culture, seems to be pure agar, but might have a purity difference to bacteriological agar. Would there be any issues for using this for fungi at all?

I just learned to fight condensation in a dry climate, I can't imagine working with humidity in PR.

My secret is while my agar is cooling allow the plates to cool face down on a rack above the bottom of a SAB, or I guess those lucky with flowhoods you can have it blow over the plate.

In the Sab i’d cover arm holes and let the plates sit for a few days unwrapped. With the flowhood a few hours in front the flow hood. Sometimes I’ll get a little fog but clears up quick. Lab RH about 66% since I share it with fruiting space.

I just went thru experimenting diff ways for pre-pour, terrible.

So I'm back to pouring in a SAB, 113F, possibly stacked with a sleeve over and left for a day or so.
They usually come out decent except for the top ones.
But it seems with temp swings from 78-85F it creates condensation.
Lately I've been covering the plates with printer paper, ok...
Next I might try a large cooler to stabilize temp.
Oh, and a dresser drawer might work too. I've been keep plates there that aren't inoculated.
Course I have about a 10% edge contam rate there, but that's another story...
