I just received some petri dishes and I couldn't find any concise information on how to manage the lids.

Some resources recommend using cling wrap or plastic wrap around them whereas some suggest that nothing should be required and the lid in itself is okay.

Any advice or direction is very much appreciated.

Oh good it’s not just me.
I swiped a print labeled playa u elf, which just meant it came from a certain cap pheno that’s pretty cool looking.

Mines doing something similar, growing under the agar mostly and having a few strands pop up here and there. I finally just dropped a plate to grain last week. It’s almost done but...still not sure what’s up lol. Suuuuper thin growth to the point I’m not sure it’s myc, but then it’ll hit a sunflower seed and look normal for a stretch.

I hit Nichrome up about it but didn’t hear back yet so idk if it’s common.



Yeah, same giveaway where my print originated. I'm getting top agar growth but a bit fluffy except for the dip in ring around the center.

Curious to hear about what happens with your grain and what Nichrome says about it. Maybe it's typical of PU Elf, albeit strange looking mycelium. Thanks for sharing, good to know I didn't mess up somewhere else.

Yeah it looks weird but grows fine ime so far.

I’ve finally got a solid ms and clone culture now for playa that looks normal.

I use a little piece of this stuff that you put underneath rugs so that they dont slide on hard floors. It's similar to shower mat kinda.

I just spray the bottom of my SAB with a little water before I put this down. Then I can lift the lid with one hand, make a one handed cut with my other and then put the lid back on once I lift the wedge out.

Cling wrap works great, cut a 1" piece off the roll to make a mini roll and its perfect. guess you could use nothing but those lids come of pretty easy

You definitely need to wrap dishes. You can keep them unwrapped until they're inoculated if they're back in the original sleeve or in a large ziploc or whatever but if they are sitting out in the open air they need to be wrapped or you will start seeing mold pop up near the edges.

I have a bread knife that works great. Also you can cut width to do a stack of five for storing.
There's a video somewhere wrapping a stack...

A rack with a little grip would be awesome. I usually just use a petting dish (empty) below the Other dishes I’m using.

Take some from all the ones on the left, that top one looks good for sure and the others are fine.. I’m no agar pro but I’ve taken cultures from ones similar to what you have and they turn out good

Has anyone tried straining agar sediments with coffee filters? Does it work? Going to the store today so I might pickup a cheesecloth if coffee filters doesnt work

What kind of agar? I get little to no sediment in my plates with lme. Also once agar is PC I don’t swirl it or anything and leave the last 1/4 in the media bottle. That way any sediment that may be present stays in the bottom and not on a plate

Coffee filter didn't work for me,
neither did our cheesecloth tea strainer, which is prolly a tighter knit than standard cheese cloth.
Checkout c10 for cheese cloth reqs.

I'm about to put wedges of these plates to rye-- can I get any advice on sectors you'd choose over others. They're not too big, at 10 days from transfer, but I want to stay ahead of contamination (one of the SAT's already has competition on the far left)
