You Shirley can.
1 drop should be enough. Streak with a flamed loop or scalpel.

So how does this look? It was put in the fridge clean and pulled out just 24 hours ago. Can I hit grain with it ? Sorry about all the condensation.

Sorry to derail the thread again. Do you have to use the whole dish to inoculate or can you use a section? If there is a thread thread that already covers this I would love to know!

You can use just a section, but the entire agar plate still needs to be clean of any contaminate.  The more sections you use give the grains more contact area and you will get quicker colonization (see Tiger Drop method). 

Some people believe you should not use the entire plate as there is a greater likelihood that there are contaminates on the outer edge where the condensate drops run off the container walls.

It only take a a microscopic bit of culture to inoculate with: the more culture used, the faster the growth and the lower the risk of contamination: IF THE STARTING CULTURE IS CLEAN

I know it's been asked but gonna make grain water agar. Do I just use half grain water half water? I thought about all grain water but that may be too much. Thanks guys

Depends on how long the "soak"... If its just ran over the grain, undiluted works... If it sat for 24hrs, 1:4 prolly a better utilization IMO

Also, the temp of the water when added: hot water will be more effective at extracting nutrients..

It will be boiling water, add grain, take off heat and 45 min soak.

150ml grain water to 350ml h2o works pretty good. I did 250ml grain water to 250ml h2o and the cultures were slower. Thats with boiled grain.

Optimum grain soak water ratio is really a trial+error thing because it depends on how much water you used to soak the grains (the grain:water ratio in the soak/boil), how long they were soaked, etc. IMO a good starting point to try is 25% GSW, it might not be perfect but it most likely work.

Here is an update.... from complete failure and contamination to this.

I have a bunch of agar going... but one of the plates that got started August 15 with MS syringe has blued, and now pinned into nice visible pins as opposed to tiny white dots, though those are there as well.

I'm happy that I failed hard initially and this turned me to agar.  I'm sort of dreaming of a stereo microscope... This is awful!



Should I wait for the pin to develop more and then clone or should I clone right away?

Clone the pin! Do some transfers....this plate still has potential to give you some good myc to work with. :super:

i filter my gw for this and don't dilute it. but like said it's gonna depend on a few other factors, so when you got your grain prep consistent, it's easy to test different dilutions and see what you like.

1.  Do I need to take any additional precautions when cloning the pin?
1a. Can i consider it clean and not have to cut into it [or rip into pieces]?  It's small to destroy like that.

I do have other plates going from this culture I'm on transfer 4 right now.

This is T2 Aug 23, look at that primordium.  Not very well focused due to condensation on the lid. 



2. Should I also do more  transfers of the one above? Since it is forming lots of bodies even on agar.



Here is the most recent T4 done 2 days ago.  Please note that the 5 o'clock is clean, it's something on the outside.



3. If agar transfer is successful  should i do  Agar to Grain, and then G2G? 

4. What's a good way to store the culture on that plate that is currently fruiting?

I made the Grain water agar, I went 50/50 and it SUXXXXXX. I transferred from beautiful mycelium, and the new plates have grown but the mycelium is very faint. Should I have used more or less grain water? WTH
