How does this clone from a wild Cyclocybe parasitica look? do I need to make more transfers? It looks clean to me.


If that is sporulating mold, probably not. If its bacteria, then maybe. I cant tell from the pic

Also, i do this with clones/spore plates: I always do at least one transfer, just so im not inoculating with the original clone/spores (which may be contaminated) and making sure theres no latent contamination. I usually clone and isolate on MEA and then switch to PDA for the inoculation plates.

Always always ALWAYS wait to use a culture until the dish is clean ENTIRELY... You're dealing with micro-organisms... smaller than the eye can see... if you see it, it's likely EVERYWHERE  in that dish... transfer, invest an extra plate and day or 3... save yourself weeks, and more resources on the back end trying to cut corners now

some put in fridge. I just put them back in the sleeve with a zip tie at room temp. any bad plates will show mold after couple days this way so no surprises.

PDA is a preference. try different things, see if you like one more. I only use GWA from beginning to end and have no issues.

So, sure it's a preference thing, but i'm curious as to what is the reasoning behind the preference.  If it is was simple:  "i like MEA" then one would use MEA for innoc too, not switch to PDA.  This raises questions like,  does it colonize faster?  or perhaps it is something else.

Just like you eating the same thing day in day out, it can get boring. It just gives the myc a bit of variety, keeps it happy. I couldn't be annoyed myself and just use grain water with a bit of lme for everything.

Ok I'll make some transfers today. All my plates are like 3 months old so there is no leading edge to transfer from, should I just pick a healthy looking piece from the middle

Wow... good luck... any contamination present is likely meshed with the culture at this point and may NEVER come clean...

I store unused, unwrapped plates in gallon Ziploc for upto a month sometimes with no issue... thx Mycolorado for the turn on (I'm pretty sure?) Been doing it since going laminar and never had any issues

Have some faith :lol: they all showed nice uniform growth with no signs of contams for at least a month. The contamination is only on the sides/rim of the petri and the mycelium seemed to smoother it pretty quickly.

Not too concerned if it ends up they're screwed they've survived a lot of rough handling and big temp fluctuations so anything I can get from them is a bonus

If you're that confident, why transfer at all? I mean, take one for propagation if you don't keep masters, but a clean culture is a clean culture... use it :shrug:

until they die or get contaminated. With a fridge you could be talking years. Got more specifics on why?

until the plate pins and that pin sporulates, and probably longer after that. I wouldnt use it if it got spored on, but someone might. As long as it doesnt contaminate or die back, it's viable. Some cultures will run their entire life cycle on a plate, if neglected long enough.

Iv kept plates in the fridge for 6 months and they worked fine when dropped in grain.

Edit so I am not double posting: I thought I saw something that said you could put spores from a syringe onto the agar as well. Is it a matter of just sterilizing the needle and doing a couple drops into the agar?
