I added some rye flour to my last batch of agar. Busted out another APE swab to see what results and started some Corumba Brazil as well.

What do you guys do with old plates? What about ones that pin? I was always told not to throw em out but once they fruit then what?
The plate is a clone plate, so does the fruit on the plate have any more desirable qualities than the clone culture? Or is it just a great sign  that the culture is good?

A clone will still contain multiple strains, taking an agar pin clone further isolates easy/known fruiting strains IMO... just depends on how close to isolate you are whether it'll help any, but won't hurt

I don't think it will affect genetic composition of culture in noticeable degree. Eg. if you have two strains in single mushroom, one with 105% growth speed of second one (let's call better strain A and worse strain B), "first generation clone" will be 51.2%A, "second generation clone" will be 53.7% A, "third generation clone" will be 59% A etc.

But there is something that can be done! This fruiting body was grown in sterile container, so you can blend it in sterile blender, dilute liquid and then perform streaking. If your inoculant will be diluted enough, you should be able to find fast growing strains.

I need help!

I might just be dumb but I cannot find a good small batch agar recipe and not sure how to scale down the ratios. My agar usually is not super clear. If I use too little agar it is runny and hard af to do transfers. If I put more it’s cloudy and sediment is visible.

For this batch I was aiming for 8 mini rounds. I measured by filling 8 mini rounds around the wave lines at the bottom of the cup. It came out to a little over half a cup of water. I added 1/8 tsp of brf which would not disolve even when heated. I added 1 tbs of agar, let it cool to test consistency and it was runny so I added a tsp of agar more.

Not sure if this is the wrong kind of agar??

I’ve got by with sub par agar but the growth is usually tomatoes and only ever see rizo after a few weeks of the plate getting overgrown. Still able to get agar to grain and get decent flushes I am just aiming to level up. Thanks for reading and any helpful tips! :smile:

How long can you keep agar plates before they start pinning? Also not sure if pinning is a guarantee of that is more of an oddity.

To scale down just divide the amount of each ingredient to your desired level. The standard recipe i use is 20g agar: 20 g lme: 1 liter (1000ml) water. I usually only want to pour 10 plates so I'll scale it down to 6.7g agar: 6.7g lme: 333 water.
That agar works well but it takes a lot of heating and stirring to fully dissolve. The "Now" brand agar dissolves alot quicker since it's powdered but seems to make a firmer agar ime.

My isolates are a result of propagation, that I happen to keep track of over time: not something I'm doing on purpose... the sectors of growth that are visually ahead of the other sectors are generally safe to be assume as the faster growing no?  This percentage jazz and dillution/pollution? Too much man... if it's not apparent to the naked eye on the plate that it's faster  (not necessarily what you're looking for)

As far as cloning a clone: advantages stand the same as cloning a fruit from MS... pick the fastest and largest finishing fruit from a cluster and continue to thin out the fast, large, clustering genes... or make a squirt gun and end up with someone else genetics...

Your not seeing only 3 strains in an MS fruit clone culture... you MIGHT get lucky and see ONLY 300...

Instead of piling into this thread with my cleanup attempts, i split out into a separate one. I have lots of questions, but i would love even just have one or two of those answered.
In short I need some help assessing if I'm growing anything useful, or if this is all mold and bacteria.

In order to stay on topic here is another plate I have after 1 transfer from  spores (due to condensation I stored it upside down, but decided to shake the droplets off today.)
Should I do another transfer or does that look ok?

I do have another transfer going, but the condensation is killing me...

Standard MEA should work for anything... shit will grow a clone of granny's bunion!

Nomad, not entirely sure that's even cubensis...

Like mold iffy... like I'm almost more sure it's unintended mold more than cube myc... idk though: one transfer from spore, may just be puffy from latent bacteria... I'd continue to transfer at least T3 before considering use personally... rather invest a few extra days and a few extra plates to be sure,  than lose weeks, grain, coir etc

I suggest taking transfers from sections of the leading edge that appear different, and noting which perform best... this will help train your eye for what to look for: both in a good, and a bad culture

Seems like alot of nutrients, which isnt a problem but how much is it? I bought premixed mea for a while until i realized it was cheaper to buy a pound each of plain agar and light malt extract. 20 dollars for a years worth of agar, and add 60 for the 500 case of petris and agar work is paid for for at least half a year, if youre being slightly conservative with transfers.

Also nomad id take a transfer from both the center and the edge and compare. The first two pics just look like alot of isolation is needed, the third one looks bad.

did u already buy it? if not, go with landor enterprises. MEA, 25 g for 7.99 i think. Just type in landor enterprises on amazon. Itll work just fine, theres just better options

I would spend that 10 on just malt extract and go to an asian grocer and buy good ol telephone agar agar powder.

Could I cut the contamination out of these oyster plates and use them to inoculate a saw dust block or should I make some more transfers? I think it's pretty cool how the mycelium seemed to attack the contams.



How does this clone from a wild Cyclocybe parasitica look? do I need to make more transfers? It looks clean to me.
