I've got some nice looking plates coming along that were on WO agar.  Question is should T1 go to WO also or should it be more nutritious?

Freezing does modify starch… a basic web search will show this. Mycelium process digestible and resistant starches differently.

If you have such a germination plate, do you wait for mycelium to grow bigger, or would you make transfers now, each spore clump to the new plate?

Personally I'd split them up. Each distinct culture will have their own performance, I'd prefer to let them develop independently / without competition.

This is a transfer from a germ plate; it has a brother from the same time. I did a transfer from brother, and I swear got a MOLD scent from outside the SAB. Sharp like white mold on an orange. So I opened this one and sniffed it a week ago. A day later had Mrs. Bluescreen sniff it. No mold smell, nice smell. It was around the ridge visible at that point, been unwrapped since then, and fully grown out since. Crazy. Maybe it's fuckin mold anyway 'cause I sent another transfer to grains when this plate was done and ain't got jack growing.



Edit- I opened that jar as the grains had gone a lil mushy upon shaking, cautious sniff and it smelled like cooked grains. Gone now.

If you have a 100mm plate fully colonized, how many grain jars would you (not could you) typically inoculate with that 1 plate?

4-8 but I only do 1 wedge to each jar because I don't trust myself :lol:

For the most part I do 4 jars from the same 55mm plate and I don't let my plates get 100% colonized as that has the small chance of a contamination from the gap just going dormant on top of the mycelium and when it hits the grains, germinate, but I'm paranoid :uhoh:

This is ochra myc on a plate; these spots appeared in the last few days- wtf are they?! They are like dark water droplets but a bit translucent up close with a loupe. I'll keep it around until it does something (hoping it's just ochra being weird and that shit pins :headbang:)

Second pic is with a loupe at the camera lens, a little better close up shot. Backside looks completely normal.

It kind of looks like it's trying to pin on the plate, I also found growing Ochra's that you may see more metabolites than you are used to, I'm seeing a bit on the Ingeli tub as well.

Okay, so this is my first time working with agar, and that being the case, I'm still not really sure what things should be looking like. I have a feeling I'm cultivating mold here, there's a weird transparent leading edge on the growth that seems sus, but I've also never seen mycelium from freshly germinated spores on agar before and I'm not familiar with how it behaves yet. These plates have slices of spore swabs on them lol, not sure if thats common practice, I'm sure it's risky, but I swabbed a few plates and wasn't seeing much spore transfer so I just tossed a chunk on another because why not. Anyway, I guess that's just a long winded way of asking, do these look fucked? Thanks

Might be mold, what species are you trying to grow?

It is bacterial anyway, that's for sure. You'll want to cabin sequester that. Unless of course you've got more plates, or more prints / swabs to work with. Cleaning up things is something to avoid if you can.

They're cubes, acadian coast and brainy shakti. I anticipated having to do multiple transfers and have more plates poured and ready to go, as well as stuff for another pour, so thats not an issue. The plates I streaked have still yet to germinate, save for one that's def growing a contam. The plates I put slices of swab on were a bit of an experiment/hail mary incase the streaked ones didn't work for whatever reason. I'm just gonna let them grow out more and see what they end up looking like.

I think I just need to be more patient and wait for the streaked plates to germinate. I have a scope(only 40x though) and was able to confirm that the plates that initially looked like nothing was on them do in fact have spores on them. Except for the shakti, it's albino and I dont think my dinky scope can make out the light colored spores or something.

Should note that I have several grows under my belt already, this is just my initial foray into agar work, no more ms to grain for this guy 😅

What is the agar recipe? Bacteria are usually white or orange. The agar looks soft and/or watery. Try leaving your plates standing upright, if it is bacteria it will follow gravity while the mycelium can escape upwards.
