Why my Yeti isn´t expanding on petri plate?

I transferred from WA to PDA, i did 3 plates, 2 contaminated 1 clean, but the clean one isn´t expanding.

The transfer date was 01/mar.

PD: The small dots, marks, or stains are from outside the petri dish; inside, the agar looks excellent.

Give it another week mate, the fact it hasn't exploded with bacterial shit means it should be clean. Make sure it's wrapped and kept between 72-76 if you want best results. I've had something not moved for 7-8 days after transfer and then explode.

:agar: really is an art unto itself, you'll get to know each of your cultures and how they behave - if you pay attention. I'm only a few months in and I consider every culture an individual soul with their own likes/dislikes, appearances, and growth patterns.

Good luck, god bless, dont give up!

Good morning guys Nd' gals, I hope the morning is treating you kindly...

So I'm wondering are their any constraints to what I can recycle and use to make agar in to then pour? Like for example...

Could I half fill (1L) a couple and stack em in the PC to get 3L of agar in one go dya think?

Or alternatively would a bunch of modded grain jars filled with it just be the one, I realise I need to get a big LC bottle but I'm a broke till next week and I have a ton of things need doing.

Be happy, stay healthy shroomers.

You could, but it would be a mess to pour. Media bottles work great because they have a ring that prevents spilling and whatever you're pouring, running down the side of the bottle.

I'm not sure I could ever go back to pouring agar without a media bottle. :lol:

Anything glass and/or polypropylene you're comfortable pouring out of, but IMO things with narrow mouths work best. A mason jar is probably easier to work with than the plastic container pictured. An old liquor bottle or narrow vase would be even better. If you don't have a lid or stopper, you can use a wad of polyfil and cover with foil to keep the lip sterile.

Or you could pre-pour your plates if they're not the disposable kind.

I’ve got several media bottles and a nice Pyrex Erlenmeyer flask. They work fine, but my preference is a 1L PP graduated pitcher from Rural King or Tractor Supply. I don’t recall which. I have a box of aluminum foil sheets from Sam’s Club that secure over the top, and can be folded back to expose the pour spout only.

As purpose built as the pitcher is, I still manage to dribble occasionally or get too confident and splash pour a plate.

I cannot imagine trying to pour from a leftovers container.

3L? 1 SAB session?

:goodday: Thanks for the answers you sacred people.

So what I'm gonna do is no pour again and use these:

Once I finish making em up, the center is clear and sides taller for a much better view. Media bottle is first thing I'm ordering Tuesday!

I want everybody here at the shroomery to know I really really just wanna help where I can help and be around people with higher thinking, like if I can't find my place here, I'm a bit scared, because this is the my last stop with humanity - the last corner I'm looking in before I become a complete hermit.

The only way I reach out anymore is through here. I talk to 0 people outside of the shroomery besides my girlfriend (haven't physically seen her in 4 months either).

I think I'm tryna say this: I think I get how off and odd I come across - though there's a load I don't understand, but I come from a genuine place with only good intentions.

Please have patience with me, I'm not a normal person strutting into the community, I'm damaged beyond damaged and I get we all are yeah yeah - but this is different - despite how capable I might come across.

Nd' if I'm out of line please say something to me, I'm up to 5 people blocking me now and not a hint as to why :woooaaahhh:

Thank you for your time, mush love xXxX:mushroom2:

For cubes, do yall like to use MEA or PDA?

I used PDA exclusively for years, now I'm going MEA mostly for ease of use.

Whats in your dish?

How's my agar look?

These 2 were inoculated with spores on the 2nd of March.

I used BRF as the food in my agar, so they are a little cloudy :/

@Peleus

Splittinghairs and Phaethon are both right haha, I HATE trying to judge plates online. You've got some nice even growth, but on photo it can be tough to impossible to judg the organisation of the leading edge.  What can be simply be faint, new myc can look on photo as a greasy halo of a ride along/competitor of some sort. And vice versa...

That is one of your biggest clues, the edge and its level of organisation.
Although the community can help, theres no substitute for observation and getting to know your own cultures IMO.

Taking a T3 but keeping the donor for observation is good here I reckon, if for nothing more than education.
I like having a little jewellers loup around for my mushroom stuff, it can be handy to study your plates edge with too.
:idea:
@PortabellaKindaguy,
went the other way than yourself, tryin potato after years of mainly LME use.
But I'm also very fond of using nice murky grainboil water agar.

@ jefferykite looks like you got something to work with!

Thanks Lithop, I'm actively trying to learn how to read plates and the magnifying glass is a great idea!
On these particular plates, the transfers were taken from the leading edge, well before the edge mold showed up. I'll watch all plates closely and take another transfer to be sure.

What's your grainwater recipe? I'm currently on standard 2% LME, 2%agar and was thinking of using that grainwater but dunno if I should cut down on the LME or keep everything as is.

Sorry. Was on vacation so my response wa quick.

Lit pretty much said what I was thinking.  I’m seeing organized rope like growth.

I (personally) wouldn’t be sold on how light the photo makes it look. And if I read your labeling correctly it’s golden teacher.  So I would go one more just to make sure something ain’t riding along which with the thin look to it is what I am concerned about.

But like Fit said, take a transfer and let the old grow out.  I always do that also

:yesnod:^

@Peleus- Good luck with it!

Done some transfers last night and I'm trying to not go and inspect the 0% of progress:lol:
My eye is practically twitching.

As for the grain water agar, I simply keep 250ml of the water I cook my grains in then use it for the agar.
