Re: AGAR ENVY! (Anything and All things agar!) [Re: ModularMind]

I'm seeing the same behavior on my Eryngii plates. They are also clones, so maybe the mycelium stayed in "fruiting mode" instead of reverting back to colonization? I mean, it's colonizing the plate fine, but it started off with fruiting. I really don't know why. The substrate is PDA with a little baking yeast in it.

I also have maitake that I’ve transferred away from a biopsy plate. There was bacteria near the original clone pieces, but the leading edge looked alright. Haven’t seen any initial growth on those transfers yet.

Thanks. I was excited when I saw it.

Can anybody help me with my water agar plates, please?
I keep getting those yellowish rings and spots.
Not sure if it's bacteria, mold, yeast or even something normal.

The species are:

Ps. cubensis Fidgi
Pan. cyan. Hawai
A stoneproducer cloned from Dutch sclerotia (Valhalla)

Already tried BRF pucks, cabin sequestering and water agar again and again.

I'm starting to despair.
Thanks in advance for some input what this yellow stuff could be and how to combat it.

Those all look clean to me aside from maybe the second one F-WA.

The indication is that the yellow ring is a ring. i.e. organized.  This suggests a metabolic shift due to age, nutrients, and how much of the plate the vegetative mycelium has captured.

F-WA the ring is not well as organized, which is a bit suspicious.  But considering it's been cabined, ran through water agar, etc.  I would take it back to a nutrient plate and post a picture when it grows out a bit.  I imagine it's fine too.

I have a bunch of 3 cell plates. It would be neat to have an army of cordoned off… incels?

Old coffee bottles with black agar cooled on its side.
The one was cooled while spinning on a freezer ice pack, and is being colonized from the bottom up around the entire inside. Just for fun and decoration.

So my homemade tubs with a hint of peptone (all in fridge cept' pan india)-

TTBVI (bit concerned bout that colour! Looks blue but...)

Tampanensis - starting to form sclerotia I think, in a fridge!

B-bar

Nec-D

Eungai

And Pan India which hasn't been assigned yet, it's closely related to pan cyan apparently, tree frog has the deets on it. It went contam but it's from spores so ya know. It's got a nice edge their I'll take a few transfers from and cabin in 0.5% LME agar. No more peptone! These are all 1% LME .5% peptone. Peptone just encourages bacterial bs.

I pray everybody's agar dreams come true!

Does this look right to you guys?

It's an A+ cube, but the mycelium is so much fluffier compared to other cubes I've got. Based on colonisation speed I doubt it's mold.

You camera is focused too close and the cover is more clear than the culture, but it looks alright to me. Maybe transfer again to a lower nutrient solution? What are you running it on?

Hi Arche, sorry about the picture, it's the best I could get, always a struggle to get focus to land on myc.
It's a standard 2% agar, 2% lme recipe.

I guess I won't know for sure until I send it to grains. This is already the 4th transfer.

Brother it's looking great, rhizo growth is super optional, not even optimal in all scenarios.

You're making solid progress, it'll be second nature before you know it :fingerpistol:

Thank you for your answer, tree frog.
That calms me down a lot.
Will follow your advice and transfer back to nutrient plates and post updated pics.
Thanks for your input. :sun:

Why my Yeti isn´t expanding on petri plate?

I transferred from WA to PDA, i did 3 plates, 2 contaminated 1 clean, but the clean one isn´t expanding.

The transfer date was 01/mar.

PD: The small dots, marks, or stains are from outside the petri dish; inside, the agar looks excellent.
